  • The Bovine CCL4 (MIP-1 beta) ELISA is a sensitive immunoassay designed for the quantitative detection of bovine CCL4 (macrophage inflammatory protein-1 beta) in cell culture supernatants. With minor protocol adjustments, this ELISA kit can also be used to measure CCL4 in additional biological sample types, offering flexibility for a wide range of immunology, inflammation, and disease research applications. The assay exhibits cross-reactivity with Yak, American Buffalo, and Zebu CCL4, making it an effective tool for comparative immunology and translational studies across related species. Bovine CCL4 (MIP-1 beta) has been successfully detected in multiple sample matrices, including cell culture supernatant, plasma, serum, lymph nodes, gingival tissue, and bronchoalveolar lavage fluid. As a key pro-inflammatory chemokine, MIP-1 beta plays a critical role in immune cell recruitment, particularly T cells and monocytes, and contributes to immune regulation and inflammatory signaling pathways. Quantifying CCL4 levels helps researchers better understand immune responses, inflammatory disease mechanisms, and disease progression. Elevated MIP-1 beta expression has been associated with conditions such as HIV infection, multiple myeloma, multiple sclerosis, and cancer, where it may serve as a biomarker for inflammation severity, bone resorption, and antiviral immune activity. The Bovine CCL4 ELISA kit contains a capture antibody, detection antibody, and a recombinant CCL4 protein standard for assay development. These antibodies have been validated to work together in an ELISA format using the provided standard. Assay conditions-including buffers, antibody concentrations, incubation times, and temperatures-have not been pre-optimized, allowing investigators to tailor assay performance to achieve optimal sensitivity and dynamic range for their specific sample types. Kit component quantities are not matched, and individual reagents are available for purchase separately. This product is for research use only and is manufactured in the USA.

Bovine CCL4 (C-C motif chemokine ligand 4, also known as MIP-1β, macrophage inflammatory protein-1 beta) is a pro-inflammatory chemokine that regulates recruitment and activation of immune cells during inflammatory and infectious responses in cattle (Bos taurus). CCL4 primarily signals through the CCR5 receptor, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection or tissue injury. In cattle, CCL4 is produced by activated macrophages, dendritic cells, epithelial cells, and lymphocytes in response to inflammatory stimuli such as microbial components and cytokines including TNF-α and IL-1β. CCL4-mediated immune cell recruitment plays an important role in host defense against pathogens involved in diseases such as bovine respiratory disease complex (BRDC), mastitis, and mycobacterial infections including Mycobacterium bovis. In these conditions, infiltration of monocytes and lymphocytes contributes to pathogen clearance and activation of adaptive immune responses. However, excessive or prolonged expression of CCL4 may contribute to inflammatory tissue damage and chronic inflammation. As a regulator of leukocyte trafficking and inflammatory signaling, bovine CCL4 is important for studies of host–pathogen interactions, inflammatory disease mechanisms, and vaccine responses aimed at improving immune protection and disease resistance in cattle.
