  • The Bovine IL-1α (Interleukin-1 Alpha) ELISA is designed for the quantitative measurement of bovine IL-1α in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-1α in additional biological sample types reported in the literature, including serum, plasma, whole blood-derived culture supernatants, peripheral blood mononuclear cell (PBMC) culture supernatants, tissue homogenates or lysates, bronchoalveolar lavage fluid (BALF), and milk, depending on the experimental model and disease context. This flexibility supports a wide range of immunology and translational research applications. Interleukin-1 alpha (IL-1α) is a pro-inflammatory cytokine that plays a central role in innate immune activation, inflammatory signaling, and host defense. Produced primarily by monocytes, macrophages, and epithelial cells, IL-1α initiates local inflammatory responses, activates endothelial cells, and promotes recruitment of immune cells to sites of infection or tissue damage. Measurement of IL-1α is widely used to assess inflammatory responses, immune activation, and disease progression in both experimental and clinical bovine models. In bovine and related species, IL-1α serves as an important biomarker of acute inflammation, infection, and tissue injury, supporting research in animal health, comparative immunology, and translational studies. The bovine IL-1α protein is 100% homologous to American Buffalo (Bubalus bubalis), Banteng (Bos javanicus), and Zebu (Bos indicus) IL-1α, making this ELISA well suited for studies involving these species and closely related animals where sequence identity supports cross-species detection. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine Interleukin-1 Alpha (IL-1α) is a pro-inflammatory cytokine belonging to the IL-1 family, which includes IL-1α, IL-1β, and the IL-1 receptor antagonist (IL-1Ra), and plays a central role in initiating and amplifying innate immune responses in cattle (Bos taurus). IL-1α is constitutively expressed in epithelial and barrier tissues and can function as an intracellular alarmin that is released upon cell injury or necrosis, triggering rapid inflammatory signaling. It binds to the IL-1 receptor type 1 (IL-1R1), activating NF-κB and MAPK pathways that promote leukocyte recruitment, endothelial activation, fever, and production of additional pro-inflammatory mediators. In bovine health, IL-1α is particularly relevant in inflammatory conditions such as mastitis, bovine respiratory disease (BRD), metritis, enteric infections, and lameness-associated tissue injury, where it contributes to early neutrophil infiltration and local inflammatory amplification. While essential for pathogen defense and tissue repair, excessive IL-1α signaling can contribute to tissue damage and reduced productivity. As an early mediator of sterile and infectious inflammation, bovine IL-1α serves as a biomarker of tissue injury and innate immune activation.
