  • The Bovine IL-1 Receptor Antagonist (IL-1RA) ELISA is designed for the quantitative measurement of bovine IL-1RA in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-1RA in additional biological sample types reported in the literature, including serum, plasma, whole blood, peripheral blood mononuclear cell (PBMC) culture supernatants, milk, colostrum, tissue homogenates or lysates, bronchoalveolar lavage fluid (BALF), synovial fluid, and cerebrospinal fluid (CSF), depending on the experimental model and disease context. This flexibility supports a wide range of immunology and translational research applications. Interleukin-1 receptor antagonist (IL-1RA) is a naturally occurring inhibitor of IL-1 signaling, binding to the IL-1 receptor without inducing downstream pro-inflammatory effects. IL-1RA plays a critical role in modulating inflammatory responses, controlling immune activation, and maintaining tissue homeostasis. Measurement of IL-1RA is widely used to evaluate anti-inflammatory regulation, immune balance, and disease progression in both experimental and clinical bovine models. In bovine and related species, IL-1RA serves as an important biomarker for inflammatory regulation, infection response, and tissue protection, supporting studies in animal health, comparative immunology, and translational research. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine Interleukin-1 Receptor Antagonist (IL-1Ra) is an anti-inflammatory cytokine produced by monocytes, macrophages, neutrophils, and other immune cells in cattle (Bos taurus) that functions as a natural regulator of IL-1–mediated inflammation. IL-1Ra competitively binds to the IL-1 receptor type 1 (IL-1R1) without triggering downstream signaling, thereby inhibiting the pro-inflammatory effects of IL-1α and IL-1β and limiting activation of NF-κB and MAPK pathways. In bovine health, IL-1Ra plays an important role in modulating inflammatory responses associated with economically significant conditions such as mastitis, bovine respiratory disease (BRD), metritis, lameness, and enteric infections, helping to balance pathogen clearance with prevention of excessive tissue damage.
