  • The Bovine IL-13 (Interleukin-13) ELISA is designed for the quantitative measurement of bovine IL-13 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-13 in additional biological sample types reported in the literature, including serum, plasma, whole blood, peripheral blood mononuclear cell (PBMC) culture supernatants, tissue homogenates or lysates, milk, colostrum, bronchoalveolar lavage fluid (BALF), and synovial fluid, depending on the experimental model and disease context. This flexibility supports a wide range of immunology and translational research applications. Interleukin-13 (IL-13) is a key Th2-type cytokine involved in the regulation of humoral immunity, allergic responses, and anti-inflammatory signaling. It is primarily produced by activated T helper 2 (Th2) cells, mast cells, and basophils, and functions to promote B-cell differentiation, antibody production, and modulation of macrophage activity. Measurement of IL-13 is widely used to assess Th2-driven immune responses, allergic inflammation, and immune regulation in bovine and related species. IL-13 serves as an important biomarker of adaptive immunity, immune polarization, and disease progression, supporting studies in animal health, comparative immunology, and translational research. The bovine IL-13 protein is 100% homologous to Water Buffalo (Bubalus bubalis) and Zebu (Bos indicus) IL-13, making this ELISA well suited for studies involving these species and closely related animals where sequence identity supports cross-species detection. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine Interleukin-13 (IL-13) is a Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells and other type 2 immune cells in cattle (Bos taurus), where it plays a central role in regulating allergic-type inflammation, mucosal immunity, and tissue remodeling. IL-13 signals through receptor complexes composed of IL-13Rα1 and IL-4Rα, activating JAK/STAT6 pathways that drive alternative macrophage activation, modulation of pro-inflammatory cytokine production, mucus secretion, and IgE-associated immune responses. In bovine health, IL-13 is particularly important in responses to parasitic infections such as gastrointestinal nematodes and lungworms, where Th2 polarization supports parasite clearance and tissue repair, but may also contribute to fibrosis or chronic inflammation if dysregulated. IL-13 is also implicated in allergic and inflammatory respiratory conditions, mastitis-associated immune modulation, and shaping the Th1/Th2 balance during infections such as bovine viral diarrhea virus (BVDV) and other pathogens.
