  • The Bovine IL-15 (Interleukin-15) ELISA is designed for the quantitative measurement of bovine IL-15 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-15 in additional biological sample types reported in the literature, including serum, plasma, whole blood, peripheral blood mononuclear cell (PBMC) culture supernatants, tissue homogenates or lysates, milk, colostrum, bronchoalveolar lavage fluid (BALF), and synovial fluid, depending on the experimental model and disease context. This flexibility supports a wide range of immunology and translational research applications. Interleukin-15 (IL-15) is a cytokine critical for innate and adaptive immunity, primarily regulating natural killer (NK) cell development, CD8+ T-cell proliferation, and memory T-cell survival. It is produced by monocytes, macrophages, and dendritic cells, and functions to enhance cytotoxic activity, stimulate immune cell proliferation, and support immune memory formation. Measurement of IL-15 is widely used to assess immune activation, antiviral responses, and cellular immunity in bovine and related species. IL-15 serves as an important biomarker of immune competence and host defense, supporting studies in animal health, infectious disease, comparative immunology, and translational research. The bovine IL-15 protein is 100% homologous to Carabao (Bubalus carabanensis), Chinese Forest Musk Deer (Moschus berezovskii), Water Buffalo (Bubalus bubalis), Yak (Bos grunniens), and Zebu (Bos indicus) IL-15, making this ELISA well suited for studies involving these species and closely related animals where sequence identity supports cross-species detection. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine Interleukin-15 (IL-15) is a cytokine that plays a key role in regulating innate and adaptive immune responses in cattle (Bos taurus). IL-15 is produced primarily by monocytes, macrophages, dendritic cells, and other non-lymphoid tissues in response to infection or inflammatory stimuli. Bovine IL-15 signals through a heterotrimeric receptor complex composed of IL-15Rα, IL-2/15Rβ (CD122), and the common γ chain (γc), activating intracellular signaling pathways including JAK1/3 and STAT3/5 that promote lymphocyte proliferation, survival, and activation. In cattle, IL-15 is particularly important for the development, maintenance, and activation of natural killer (NK) cells and memory CD8⁺ T cells, supporting cytotoxic responses against infected or abnormal cells. It also contributes to regulation of mucosal immunity and inflammatory responses during infection. Bovine IL-15 has been studied in the context of immune responses to important livestock pathogens such as Mycobacterium bovis, bovine viral diarrhea virus (BVDV), and Mycobacterium avium subsp. paratuberculosis. Due to its role in regulating cellular immunity and lymphocyte homeostasis, bovine IL-15 is widely investigated in veterinary immunology and translational research, including studies of vaccine development, host–pathogen interactions, and immune modulation relevant to cattle health and comparative biomedical research.
