  • The Bovine IL-21 (Interleukin-21) ELISA is designed for the quantitative measurement of bovine IL-21 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-21 in additional biological sample types reported in the literature, including serum, plasma, whole blood, peripheral blood mononuclear cell (PBMC) culture supernatants, tissue homogenates or lysates, milk, colostrum, bronchoalveolar lavage fluid (BALF), and synovial fluid, depending on the experimental model and disease context. This flexibility supports a wide range of immunology and translational research applications. Interleukin-21 (IL-21) is a cytokine primarily produced by activated CD4+ T cells, including T follicular helper (Tfh) cells and Th17 cells. It plays a central role in adaptive immunity, B-cell differentiation, antibody production, and regulation of T-cell and natural killer (NK) cell function. Measurement of IL-21 is widely used to assess humoral immune responses, T-cell function, and immune regulation in bovine and related species. IL-21 serves as an important biomarker of adaptive immune activation, vaccine response, and disease progression, supporting studies in animal health, infectious disease, comparative immunology, and translational research. IL-21 is a member of the common gamma-chain (γc) cytokine family, which also includes IL-2, IL-4, IL-7, IL-9, and IL-15. These cytokines share structural and functional features, including roles in lymphocyte development, proliferation, survival, and immune regulation, but IL-21 has unique functions in enhancing B-cell antibody production and modulating T-cell and NK cell activity. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine IL-21 (Interleukin-21) is a pleiotropic cytokine belonging to the common γ chain (γc) family that plays a central role in regulation of adaptive immunity in cattle (Bos taurus). IL-21 is produced primarily by activated CD4⁺ T cells, particularly T follicular helper (Tfh) cells and Th17 cells, following antigen stimulation. It signals through the IL-21 receptor (IL-21R) in association with the common γ chain, activating downstream pathways such as JAK/STAT (predominantly STAT3) to modulate gene expression in target cells. IL-21 promotes B cell proliferation, differentiation, class-switch recombination, and antibody production, supports CD8⁺ T cell cytotoxic function, and influences NK cell activity, while also contributing to regulation of Th17 responses. In healthy cattle, basal IL-21 expression is low but increases during infection, vaccination, and immune activation. Bovine IL-21 has been implicated in immune responses to pathogens such as bovine viral diarrhea virus (BVDV), bovine leukemia virus (BLV), Mycobacterium bovis, and agents of bovine respiratory disease complex. In veterinary and agricultural research, IL-21 serves as a key biomarker of T cell–dependent B cell help and humoral immunity and is studied to evaluate vaccine efficacy, host–pathogen interactions, and adaptive immune regulation in cattle.
