  • The Bovine VEGF-A (Vascular Endothelial Growth Factor A) ELISA is designed for the quantitative measurement of bovine VEGF-A in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure VEGF-A in additional biological sample types reported in the literature, including serum, plasma, whole blood, tissue homogenates or lysates, milk, colostrum, bronchoalveolar lavage fluid (BALF), and synovial fluid, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology, angiogenesis, and translational research applications. Reported measurements: VEGF-A has been detected in bovine serum (~0.2-1.5 ng/mL), plasma (~0.05-0.4 ng/mL), tissue homogenates or lysates (~0.5-3 ng/mL), milk (~0.1-0.3 ng/mL), colostrum (~0.2-0.6 ng/mL), and synovial fluid (~0.1-0.5 ng/mL), although concentrations may vary depending on health status, experimental conditions, and sample handling. Vascular Endothelial Growth Factor A (VEGF-A) is a key regulator of angiogenesis, vasculogenesis, and vascular permeability. It is primarily produced by endothelial cells, fibroblasts, macrophages, and other stromal cells in response to hypoxia or inflammatory stimuli. Measurement of VEGF-A is widely used to assess vascular growth, tissue remodeling, wound healing, tumor angiogenesis, and inflammatory responses in bovine and related species. Why measure VEGF-A: VEGF-A is a critical biomarker for vascular activation, tissue regeneration, and disease progression. VEGF-A is a member of the VEGF family of growth factors, which includes VEGF-B, VEGF-C, VEGF-D, and placental growth factor (PlGF). These growth factors share structural features and function in angiogenesis, lymphangiogenesis, and endothelial cell signaling, with VEGF-A being a primary mediator of blood vessel formation and vascular remodeling. The bovine VEGF-A protein is 100% homologous to American Buffalo (Bubalus bubalis), Banteng (Bos javanicus), Carabao (Bubalus carabanensis), Water Buffalo (Bubalus bubalis), Yak (Bos grunniens), and Zebu (Bos indicus) VEGF-A, making this ELISA well suited for studies involving these species and closely related animals where sequence identity supports cross-species detection. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Bovine Vascular Endothelial Growth Factor A (VEGF-A) is a member of the vascular endothelial growth factor (VEGF) family, which includes VEGF-A, VEGF-B, VEGF-C, VEGF-D, and placental growth factor (PlGF)—key regulators of angiogenesis and vascular development. In cattle (Bos taurus), VEGF-A is produced by multiple cell types including endothelial cells, macrophages, fibroblasts, epithelial cells, and trophoblast cells, particularly in response to hypoxia, inflammation, and tissue injury. VEGF-A exerts its biological effects mainly by binding to VEGF receptor-1 (VEGFR-1/Flt-1) and VEGF receptor-2 (VEGFR-2/KDR) on endothelial cells, activating intracellular signaling pathways such as MAPK/ERK, PI3K/AKT, and PLCγ, which promote endothelial cell proliferation, migration, vascular permeability, and formation of new blood vessels.
