  • The Canine IL-1β (Interleukin-1 beta) ELISA is designed for the quantitative measurement of canine IL-1β in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-1β in additional biological sample types reported in the literature, including serum, plasma, whole blood, peripheral blood mononuclear cell (PBMC) culture supernatants, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Canine IL-1β has been detected in serum (~20-250 pg/mL), plasma (~15-200 pg/mL), and PBMC culture supernatants (~50-600 pg/mL under stimulation conditions), although levels vary depending on inflammatory status, infection, or experimental treatment. Interleukin-1 beta (IL-1β) is a pro-inflammatory cytokine produced primarily by monocytes, macrophages, dendritic cells, and activated epithelial cells. It plays a central role in innate immune responses, inflammation, fever induction, and cytokine cascade amplification. Measurement of IL-1β is widely used to assess immune activation, inflammatory signaling, and cytokine-mediated disease processes in canine and related species. IL-1β serves as an important biomarker of acute and chronic inflammation, infection, and immune-mediated disease progression, supporting studies in veterinary immunology, comparative immunology, and translational research. Canine Interleukin-1 Beta (IL-1β) is a potent pro-inflammatory cytokine produced primarily by activated macrophages, monocytes, and dendritic cells in dogs (Canis lupus familiaris) following recognition of pathogens or tissue damage. Synthesized as an inactive precursor (pro–IL-1β), it is cleaved into its active form by caspase-1 within the inflammasome complex and subsequently binds to the IL-1 receptor (IL-1R1), activating NF-κB and MAPK signaling pathways that drive fever, leukocyte recruitment, endothelial activation, and acute-phase protein production. In canine health, IL-1β plays a central role in inflammatory and infectious conditions including bacterial pneumonia, pyometra, parvoviral enteritis, osteoarthritis, immune-mediated diseases, and sepsis, where excessive or sustained IL-1β production can contribute to tissue damage and systemic inflammation. Elevated IL-1β levels are also associated with chronic inflammatory disorders and certain cancers, reflecting its role in shaping the tumor microenvironment.
