  • The Chicken IL-12 p40 (Interleukin-12 p40) ELISA is designed for the quantitative measurement of chicken IL-12 p40 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-12 p40 in additional biological sample types reported in the literature, including serum, plasma, peripheral blood mononuclear cell (PBMC) culture supernatants, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Chicken IL-12 p40 has been detected in serum (~50-500 pg/mL), plasma (~30-400 pg/mL), and PBMC culture supernatants (~100-1000 pg/mL under stimulation conditions), although levels vary depending on immune activation, infection, or experimental treatment. Interleukin-12 p40 (IL-12 p40) is a subunit of the heterodimeric cytokines IL-12 (p70) and IL-23 (p19/p40). It is produced primarily by dendritic cells, macrophages, and activated monocytes and plays a central role in driving Th1 immune responses, promoting IFN-γ production, and activating NK cells and cytotoxic T lymphocytes. Measurement of IL-12 p40 is widely used to assess innate and adaptive immune activation, vaccine responses, infection defense, and inflammatory signaling in chicken and related species. IL-12 p40 serves as a biomarker of cell-mediated immunity and immune response intensity, supporting studies in avian immunology, comparative immunology, and translational research. Why measure IL-12 p40: Quantifying IL-12 p40 provides insight into Th1-driven immune responses, NK cell activation, and IFN-γ-mediated immunity. It is commonly used to evaluate vaccine efficacy, infection response, immune dysregulation, and inflammatory phenotypes in poultry research. IL-12 p40 is a member of the IL-12 cytokine family, which also includes IL-12 (p70), IL-23, IL-27, and IL-35, all of which contribute to immune regulation, T-cell differentiation, and cytokine-mediated signaling. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Chicken Interleukin-12/Interleukin-23 p40 (IL-12/IL-23 p40) is a cytokine subunit that plays an important role in regulating immune responses in chickens (Gallus gallus). The p40 subunit is shared by the heterodimeric cytokine IL-12, where it pairs with the p35 subunit to form functional IL-12 that is primarily produced by activated antigen-presenting cells such as macrophages and dendritic-like cells in response to microbial infection or inflammatory stimuli. Chicken IL-12/IL-23 p40 contributes to activation of natural killer (NK) cells and differentiation of T helper cells toward Th1-type immune responses, promoting the production of interferon-γ (IFN-γ) and enhancing cell-mediated immunity against intracellular pathogens. Through these mechanisms, IL-12 signaling plays a critical role in host defense against viral, bacterial, and protozoan infections in poultry. In chickens, IL-12/IL-23 p40 is particularly studied in immune responses to important avian pathogens such as Eimeria species (causative agents of coccidiosis), Salmonella spp., and avian influenza virus.
