  • The Chicken IL-22 (Interleukin-22) ELISA is designed for the quantitative measurement of chicken IL-22 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-22 in additional biological sample types reported in the literature, including serum, plasma, peripheral blood mononuclear cell (PBMC) culture supernatants, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Chicken IL-22 has been detected in serum (~20-400 pg/mL), plasma (~15-350 pg/mL), and PBMC culture supernatants (~50-700 pg/mL under stimulation conditions), although levels vary depending on immune activation, infection, or experimental treatment. Interleukin-22 (IL-22) is a cytokine primarily produced by activated CD4⁺ T cells, particularly Th17 and Th22 subsets, as well as innate lymphoid cells (ILCs). It plays a central role in mucosal immunity, epithelial barrier function, and host defense at barrier tissues, including the respiratory and gastrointestinal tracts. Measurement of IL-22 is widely used to assess barrier immune responses, cytokine signaling, and inflammation in chicken and related species. IL-22 serves as an important biomarker of immune activation, mucosal defense, and host response to infection, supporting studies in avian immunology, comparative immunology, and translational research. Why measure Chicken IL-22: Measuring chicken IL-22 provides valuable insight into mucosal immunity, epithelial barrier function, and Th17/Th22-driven immune responses. Chicken IL-22 (Interleukin-22) is a member of the IL-10 cytokine family that plays a key role in mucosal immunity and epithelial barrier protection in chickens (Gallus gallus). IL-22 is produced primarily by activated CD4⁺ T cells, including Th17-like cells, as well as by innate lymphoid cells in response to microbial infection and inflammatory stimulation. It signals through the IL-22 receptor complex (IL-22R1 and IL-10R2), which is expressed mainly on epithelial and stromal cells, activating downstream JAK/STAT pathways—particularly STAT3—to induce antimicrobial peptide production, enhance chemokine expression, promote epithelial cell proliferation, and support tissue repair. In healthy chickens, basal IL-22 expression is generally low but increases during bacterial, viral, and parasitic infections, particularly at mucosal surfaces of the respiratory and gastrointestinal tracts. Chicken IL-22 has been implicated in immune responses to pathogens such as Eimeria spp. (coccidiosis), Salmonella spp., avian influenza virus, and infectious bronchitis virus. In poultry immunology research, IL-22 serves as an important biomarker of mucosal immune activation and epithelial defense and is studied in the context of host–pathogen interactions, gut health, and vaccine-induced protective immunity in commercial and experimental poultry systems.
