  • The Equine CCL3 (Macrophage Inflammatory Protein-1α, MIP-1α) ELISA is designed for the quantitative measurement of equine CCL3 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure CCL3 in additional biological sample types reported in the literature, including serum, plasma, synovial fluid, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Equine CCL3 has been detected in serum (~30-500 pg/mL), plasma (~25-400 pg/mL), synovial fluid (~50-600 pg/mL), and PBMC culture supernatants (~100-1000 pg/mL under stimulation conditions), although levels vary depending on immune activation, infection, or experimental treatment. Macrophage Inflammatory Protein-1α (CCL3) is a chemokine primarily produced by monocytes, macrophages, dendritic cells, and activated lymphocytes. It plays a central role in recruiting monocytes, T cells, and natural killer (NK) cells to sites of inflammation, mediating immune responses and tissue remodeling. Measurement of CCL3 is widely used to assess inflammatory signaling, immune cell trafficking, and disease progression in horses and related species. CCL3 serves as an important biomarker of immune activation, inflammatory conditions, and tissue response to infection or injury, supporting studies in equine immunology, comparative immunology, and translational research. Why measure Equine CCL3: Measuring CCL3 provides critical insight into immune cell recruitment, inflammation, and immune system activation. Quantifying CCL3 helps evaluate disease progression, vaccine responses, and immunomodulatory treatments, making it a valuable tool for both fundamental and applied equine immunology research. CCL3 is a member of the CC chemokine family, which also includes CCL2, CCL4, and CCL5, all contributing to immune cell migration and inflammatory responses. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. The quantities of kit components are not matched, and individual components may be purchased separately. This product is for research use only and is manufactured in the USA.

Equine CCL3 (C-C motif chemokine ligand 3, also known as MIP-1α, macrophage inflammatory protein-1 alpha) is a pro-inflammatory chemokine that regulates recruitment and activation of immune cells during inflammatory and infectious responses in horses (Equus caballus). CCL3 primarily signals through the CCR1 and CCR5 receptors, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection or tissue injury. In horses, CCL3 is produced by activated macrophages, epithelial cells, endothelial cells, and lymphocytes in response to inflammatory stimuli such as microbial components and cytokines including TNF-α and IL-1β. CCL3-mediated immune cell recruitment contributes to host defense in conditions such as equine respiratory infections (including equine influenza and bacterial pneumonia), inflammatory airway disease, and musculoskeletal inflammation, where infiltration of monocytes and T cells supports immune activation and tissue repair. However, excessive or prolonged CCL3 expression may contribute to persistent inflammation and tissue damage in chronic inflammatory conditions such as equine asthma or joint inflammation. As a regulator of leukocyte trafficking and inflammatory signaling, equine CCL3 is important in studies of host–pathogen interactions, respiratory disease, and inflammatory mechanisms affecting equine health and performance.
