  • The Equine IL-1α (Interleukin-1 Alpha) ELISA is designed for the quantitative measurement of equine IL-1α in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-1α in additional biological sample types reported in the literature, including serum, plasma, synovial fluid, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Equine IL-1α has been detected in serum (~20-400 pg/mL), plasma (~15-350 pg/mL), synovial fluid (~50-600 pg/mL), and PBMC culture supernatants (~50-1000 pg/mL under stimulation conditions), although levels vary depending on immune activation, infection, or experimental treatment. Interleukin-1 Alpha (IL-1α) is a proinflammatory cytokine primarily produced by activated monocytes, macrophages, dendritic cells, and epithelial cells. It plays a central role in initiating and amplifying inflammatory responses, leukocyte recruitment, and immune system activation. Measurement of IL-1α is widely used to assess inflammatory signaling, immune activation, and disease progression in horses and related species. Equine Interleukin-1 Alpha (IL-1α) is a pro-inflammatory cytokine belonging to the IL-1 family—which includes IL-1α, IL-1β, and the IL-1 receptor antagonist (IL-1Ra)—and plays a pivotal role in initiating innate immune responses in horses (Equus caballus). IL-1α is constitutively expressed in epithelial and barrier tissues and acts as an intracellular alarmin that is rapidly released upon cell injury, necrosis, or mechanical stress, triggering early inflammatory signaling. Upon binding to the IL-1 receptor type 1 (IL-1R1), IL-1α activates NF-κB and MAPK pathways that promote leukocyte recruitment, endothelial activation, fever, and amplification of pro-inflammatory cytokine cascades. In equine health, IL-1α is particularly important in musculoskeletal inflammation such as osteoarthritis and synovitis, where it contributes to cartilage matrix degradation and synovial inflammation, as well as in laminitis, colitis, endotoxemia, and respiratory inflammatory conditions.
