  • The Equine IL-8 (Interleukin-8, CXCL8) ELISA is designed for the quantitative measurement of equine IL-8 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-8 in additional biological sample types reported in the literature, including serum, plasma, synovial fluid, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Equine IL-8 has been detected in serum (~40-800 pg/mL), plasma (~30-700 pg/mL), synovial fluid (~60-1000 pg/mL), and PBMC culture supernatants (~100-1500 pg/mL under stimulation conditions), although levels vary depending on immune activation, infection, or experimental treatment. Interleukin-8 (IL-8/CXCL8) is a chemokine primarily produced by activated macrophages, monocytes, endothelial cells, and fibroblasts. It functions as a potent chemoattractant for neutrophils, playing a critical role in inflammation, innate immune responses, and leukocyte recruitment. Measurement of IL-8 is widely used to assess inflammatory signaling, immune activation, and host defense mechanisms in horses and related species. Equine Interleukin-8 (IL-8), also known as CXCL8, is a potent pro-inflammatory chemokine that plays a central role in innate immune responses in horses (Equus caballus) by mediating neutrophil chemotaxis, activation, and migration to sites of infection or tissue injury. Produced by macrophages, respiratory and intestinal epithelial cells, endothelial cells, and synoviocytes in response to microbial stimuli and inflammatory cytokines, equine IL-8 signals primarily through CXCR1 and CXCR2 receptors to promote rapid neutrophil recruitment and amplification of acute inflammation. Elevated IL-8 expression is associated with common equine conditions such as recurrent airway obstruction (equine asthma), bacterial pneumonia, strangles (Streptococcus equi infection), colitis, endotoxemia, osteoarthritis, and laminitis, where excessive neutrophilic inflammation can contribute to tissue damage and disease severity.
