  • The Equine IL-15 (Interleukin-15) ELISA is designed for the quantitative measurement of equine IL-15 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure IL-15 in additional biological sample types reported in the literature, including serum, plasma, peripheral blood mononuclear cell (PBMC) culture supernatants, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Equine IL-15 has been detected in serum (~20-400 pg/mL), plasma (~15-350 pg/mL), and PBMC culture supernatants (~50-900 pg/mL under stimulation conditions), although levels vary depending on immune activation, infection, or experimental treatment. Interleukin-15 (IL-15) is a cytokine primarily produced by monocytes, macrophages, and dendritic cells. It plays a central role in the development, proliferation, and activation of natural killer (NK) cells and memory CD8⁺ T cells, coordinating innate and adaptive immune responses. Measurement of IL-15 is widely used to assess immune activation, NK and T-cell function, and immunomodulatory responses in horses and related species. IL-15 serves as an important biomarker of cell-mediated immunity and immune system activation, supporting studies in equine immunology, comparative immunology, and translational research. Why measure Equine IL-15: Measuring IL-15 provides valuable insight into NK cell and memory T-cell activity, immune activation, and cell-mediated immunity. Quantifying IL-15 helps evaluate vaccine responses, infection-induced immune modulation, and immunotherapeutic interventions, making it a key tool for both fundamental and applied equine immunology research. Equine Interleukin-15 (IL-15) is a cytokine that plays an important role in regulating innate and adaptive immune responses in horses (Equus caballus). IL-15 is produced by a variety of cell types, including monocytes, macrophages, dendritic cells, and epithelial cells, in response to infection or inflammatory stimuli. Equine IL-15 signals through a receptor complex composed of IL-15Rα together with the shared IL-2/15Rβ (CD122) and common γ chain (γc), activating intracellular signaling pathways such as JAK–STAT that promote lymphocyte proliferation, survival, and activation. In horses, IL-15 is particularly important for the development, maintenance, and activation of natural killer (NK) cells and memory CD8⁺ T cells, supporting cytotoxic immune responses against infected or abnormal cells. It also contributes to regulation of mucosal and inflammatory immune responses in tissues such as the respiratory and gastrointestinal tracts. In cell culture, recombinant equine IL-15 is commonly used to stimulate proliferation and survival of NK cells and cytotoxic T lymphocytes, maintain memory CD8⁺ T cell populations, and enhance cytotoxic activity in in vitro immune assays.
