  • The Feline TNF-α (Tumor Necrosis Factor Alpha) ELISA is designed for the quantitative measurement of feline TNF-α in cell culture supernatants. With minor modifications to the assay procedure, this ELISA may also be used to measure TNF-α in additional biological sample types reported in the literature, including serum, plasma, synovial fluid, and tissue homogenates or lysates, depending on the experimental model and physiological context. This flexibility supports a wide range of immunology and translational research applications. Reported measurements: Feline TNF-α has been detected in serum (~20-800 pg/mL), plasma (~15-700 pg/mL), synovial fluid (~50-1000 pg/mL), and PBMC culture supernatants (~100-1500 pg/mL under stimulation conditions), although levels vary depending on immune activation, infection, or experimental treatment. Tumor Necrosis Factor Alpha (TNF-α) is a proinflammatory cytokine primarily produced by activated macrophages, monocytes, and T cells. It plays a central role in inflammatory signaling, immune system activation, apoptosis, and host defense. Measurement of TNF-α is widely used to assess inflammation, immune responses, and disease progression in cats and related species. TNF-α serves as an important biomarker of acute and chronic inflammation, immune system activation, and tissue pathology, supporting studies in feline immunology, comparative immunology, and translational research. Feline TNF-α is 100% homologous to TNF-α from Prionailurus bengalensis (leopard cat) and Prionailurus viverrinus (fishing cat), making this ELISA suitable for comparative immunology studies across these Felidae species. Feline Tumor Necrosis Factor Alpha (TNF-α) (also known as TNF or cachectin) is a proinflammatory cytokine that plays a central role in innate immune responses, inflammation, and host defense against pathogens in cats (Felis catus). TNF-α is a member of the tumor necrosis factor (TNF) superfamily, which includes related cytokines such as TNF-β (lymphotoxin-α), Fas ligand (FasL), CD40 ligand (CD40L), and TRAIL, molecules that regulate immune signaling, apoptosis, and inflammatory processes. In cats, TNF-α is primarily produced by activated macrophages, monocytes, dendritic cells, and T lymphocytes in response to infection, tissue injury, or inflammatory stimuli. TNF-α exerts its biological effects by binding to TNF receptors TNFR1 (p55) and TNFR2 (p75), activating intracellular signaling pathways including NF-κB, MAPK, and caspase-mediated pathways, which regulate cytokine production, immune cell activation, apoptosis, and inflammatory responses. In feline health, TNF-α is involved in immune responses to viral infections such as feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV), as well as bacterial infections, inflammatory diseases, and cancer.
