  • The Mouse APRIL (A Proliferation-Inducing Ligand, TNFSF13) ELISA is designed for the quantitative measurement of mouse APRIL in cell culture supernatants. With minor modifications to the assay procedure, this ELISA can also be used to measure APRIL in additional biological sample types reported in the literature, including serum, plasma, and tissue lysates, providing flexibility for a wide range of immunology and translational research applications. APRIL is a member of the tumor necrosis factor (TNF) superfamily and plays a critical role in B cell biology, including B cell survival, proliferation, class-switch recombination, and plasma cell maintenance. Through interactions with the receptors TACI and BCMA, APRIL contributes to humoral immune responses and long-lived antibody production. In mouse models, APRIL is widely studied in the context of autoimmune disease, chronic inflammation, cancer, and vaccine response, where dysregulated APRIL signaling is associated with aberrant B cell activation and pathogenic antibody production. Measurement of APRIL provides valuable insight into B cell-driven immune mechanisms and therapeutic modulation of humoral immunity. Reported measurements of mouse APRIL have detected low picogram per milliliter concentrations in serum and plasma under steady-state conditions, with elevated levels observed following immune activation, inflammatory challenge, or disease progression, as well as measurable APRIL expression in tissue lysates from lymphoid organs such as spleen and lymph nodes. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing investigators to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Mouse APRIL (A Proliferation-Inducing Ligand), also known as TNFSF13, is a member of the tumor necrosis factor (TNF) superfamily and plays a key role in B cell biology and humoral immunity. APRIL is produced primarily by myeloid cells such as monocytes, macrophages, and dendritic cells, as well as by some epithelial and stromal cells, and it signals through the receptors TACI (TNFRSF13B) and BCMA (TNFRSF17) to promote B cell survival, plasma cell maintenance, and immunoglobulin class switching. In mice, APRIL contributes to the regulation of antibody production, long-lived plasma cell persistence in the bone marrow, and mucosal IgA responses. Dysregulated APRIL expression has been associated with autoimmune disease models, B cell malignancies, and chronic inflammatory conditions. In biomedical research, mouse APRIL is widely studied in models of systemic lupus erythematosus (SLE), rheumatoid arthritis, multiple myeloma, and antibody-mediated immune responses, where it serves as an important mediator of B cell–driven immunity and a potential therapeutic target for modulating pathological antibody production.
