  • The Mouse IL-17A ELISA is designed for the quantitative measurement of mouse IL-17A in cell culture supernatants. With minor modifications to the assay procedure, this ELISA can also be used to measure IL-17A in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, providing flexibility for a broad range of immunology and translational research applications. IL-17A is a signature cytokine of Th17 cells and plays a central role in pro-inflammatory immune responses, particularly at mucosal and barrier surfaces. It promotes the recruitment and activation of neutrophils and stimulates epithelial, endothelial, and stromal cells to produce chemokines, cytokines, and antimicrobial peptides. In mouse models, IL-17A is widely studied in the context of autoimmune and inflammatory diseases, infectious disease, allergy, cancer, and vaccine response, where dysregulated IL-17A signaling contributes to tissue inflammation, host defense, and immunopathology. Measurement of IL-17A is therefore a key readout for Th17 pathway activation and inflammatory immune dynamics. Reported measurements of mouse IL-17A have detected low to moderate picogram per milliliter concentrations in serum and plasma under basal conditions, with substantially elevated levels observed following immune stimulation, infection, or inflammatory challenge. Robust IL-17A expression has also been measured in tissue lysates from inflamed organs and at mucosal sites, reflecting localized Th17-driven immune activity. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize sensitivity and dynamic range based on their specific sample type and experimental requirements. Mouse IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays a critical role in host defense and immune-mediated inflammation in mice (Mus musculus). IL-17A is produced primarily by Th17 CD4⁺ T cells, as well as by γδ T cells, innate lymphoid cells (ILC3s), and other activated lymphocyte populations in response to microbial infection and inflammatory stimulation. It acts on epithelial, endothelial, and stromal cells to induce expression of pro-inflammatory cytokines (such as IL-6 and TNF-α), chemokines (including CXCL1 and CXCL2), and antimicrobial peptides, thereby promoting neutrophil recruitment and strengthening mucosal barrier immunity. In healthy mice, basal IL-17A expression is generally low but increases significantly during bacterial and fungal infections, particularly at mucosal surfaces. Dysregulated IL-17A signaling has been implicated in numerous experimental disease models, including experimental autoimmune encephalomyelitis (EAE), collagen-induced arthritis, psoriasis-like dermatitis, inflammatory bowel disease, and asthma. In biomedical research, mouse IL-17A is widely studied as a central mediator of Th17-driven immunity, neutrophilic inflammation, and autoimmune pathology, and serves as a key target for investigating immunomodulatory therapies in translational disease models.
