  • The Rabbit IL-1α (Interleukin-1 Alpha) ELISA is designed for the quantitative measurement of rabbit IL-1α in cell culture supernatants. With minor modifications to the assay procedure, this ELISA can also be used to measure IL-1α in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and inflammatory exudates, enabling broad applicability across immunology and translational research studies. IL-1α is a potent pro-inflammatory cytokine that functions as an early mediator of innate immune responses. It is produced by a variety of cell types, including macrophages, epithelial cells, and fibroblasts, and can act both as a secreted cytokine and as a cell-associated alarmin released during tissue injury or necrosis. In rabbit models, IL-1α plays a key role in initiating inflammation, promoting leukocyte recruitment, and amplifying downstream cytokine and chemokine cascades. Measurement of IL-1α provides valuable insight into acute inflammation, tissue damage, host defense mechanisms, and immune activation. Reported measurements of rabbit IL-1α have identified low picogram per milliliter concentrations in serum and plasma under basal conditions, with substantially increased levels detected following inflammatory stimulation, infection, or tissue injury. IL-1α has also been measured in tissue lysates from inflamed organs and at sites of localized immune activation, reflecting its role as an early and localized driver of inflammatory responses. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize sensitivity and dynamic range based on their specific sample type and experimental requirements. Rabbit Interleukin-1 Alpha (IL-1α) is a pro-inflammatory cytokine belonging to the IL-1 family—which includes IL-1α, IL-1β, and the IL-1 receptor antagonist (IL-1Ra)—and plays a central role in initiating innate and sterile inflammatory responses in rabbits (Oryctolagus cuniculus). IL-1α is constitutively expressed in epithelial and endothelial tissues and functions as an intracellular alarmin that is rapidly released upon cell injury, necrosis, or viral infection, triggering early inflammatory signaling independent of inflammasome processing. Upon binding to the IL-1 receptor type 1 (IL-1R1), rabbit IL-1α activates NF-κB and MAPK pathways that promote leukocyte recruitment, endothelial activation, fever, and amplification of downstream pro-inflammatory cytokine cascades. In rabbit health, IL-1α is particularly relevant in inflammatory conditions such as respiratory infections (e.g., Pasteurella multocida), viral diseases including rabbit hemorrhagic disease virus (RHDV) and myxoma virus, enteritis, and joint inflammation, where epithelial damage leads to rapid IL-1α–mediated immune activation. While essential for host defense and tissue repair, excessive IL-1α signaling may contribute to systemic inflammation and tissue pathology. As an early mediator of sterile and infectious inflammation, rabbit IL-1α serves as a biomarker of tissue injury and innate immune activation.
