  • The Rabbit IL-4 (Interleukin-4) ELISA is designed for the quantitative measurement of rabbit IL-4 in cell culture supernatants. With minor modifications to the assay procedure, this ELISA can also be used to measure IL-4 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology and translational research applications. IL-4 is a hallmark Th2 cytokine produced primarily by activated CD4⁺ T cells, mast cells, and basophils and is a key regulator of humoral and allergic immune responses. It drives Th2 cell differentiation, promotes B cell proliferation and class switching to IgE, and modulates macrophage polarization toward an alternatively activated (M2) phenotype. In rabbit models, IL-4 is widely studied in the context of allergy, parasitic infection, asthma, vaccine response, and immune regulation. Measurement of IL-4 provides critical insight into Th2-skewed immune responses and immune balance between pro-inflammatory and regulatory pathways. Reported measurements of rabbit IL-4 have detected low picogram per milliliter concentrations in serum and plasma under baseline conditions, with elevated levels observed following immune stimulation, allergic sensitization, or parasitic challenge. IL-4 has also been measured in tissue lysates and localized samples from sites of Th2-driven inflammation, reflecting localized cytokine production and immune polarization. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development. These antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize sensitivity and dynamic range based on their specific sample type and experimental requirements. Rabbit Interleukin-4 (IL-4) is a Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells, mast cells, and basophils in rabbits (Oryctolagus cuniculus), where it plays a central role in regulating humoral immunity, allergic inflammation, and alternative macrophage activation. IL-4 signals through receptor complexes containing IL-4Rα paired with the common gamma chain (γc) or IL-13Rα1, activating JAK/STAT6 pathways that promote B-cell proliferation, antibody production, IgE-associated responses, and suppression of Th1-associated cytokines such as IFN-γ. In rabbit health, IL-4 is particularly relevant in parasitic infections, allergic skin conditions, and respiratory inflammation, where Th2 polarization supports antibody-mediated defense and tissue repair but may also contribute to eosinophilic infiltration and chronic inflammation if dysregulated. Rabbits are widely used as experimental models for asthma, pulmonary inflammation, atherosclerosis, and ocular inflammatory disease, and IL-4 plays a key role in airway hyperresponsiveness and mucus production in allergic airway models.
