  • The Rat IL-17A ELISA is designed for the quantitative measurement of rat IL-17A in cell culture supernatants and can also be used to measure IL-17A in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology, inflammation, and translational research applications. IL-17A is a signature cytokine of Th17 cells and plays a central role in pro-inflammatory immune responses, particularly at mucosal and barrier surfaces, by promoting the recruitment and activation of neutrophils and stimulating epithelial, endothelial, and stromal cells to produce chemokines, cytokines, and antimicrobial peptides. In rat models, IL-17A is widely studied in the context of autoimmune and inflammatory diseases, infectious disease, allergy, cancer, and vaccine responses, where dysregulated IL-17A signaling contributes to tissue inflammation, host defense, and immunopathology. Measurement of IL-17A provides critical insight into Th17 pathway activation, inflammatory immune dynamics, and immune regulation. Reported measurements of rat IL-17A have detected low to moderate picogram per milliliter concentrations in serum and plasma under basal conditions, with substantially elevated levels observed following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed organs, reflecting localized Th17-driven immune activity. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Rat IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays a central role in host defense and immune-mediated inflammation in rats (Rattus norvegicus). IL-17A is produced primarily by Th17 CD4⁺ T cells, as well as by γδ T cells and innate lymphoid cells, in response to microbial infection and inflammatory stimulation. It acts on epithelial, endothelial, and stromal cells to induce expression of pro-inflammatory cytokines (such as IL-6 and TNF-α), chemokines (including CXCL1 and CXCL2), and antimicrobial peptides, thereby promoting neutrophil recruitment and strengthening mucosal barrier immunity. In healthy rats, basal IL-17A expression is generally low but increases during bacterial and fungal infections, autoimmune responses, and inflammatory challenge. Rat IL-17A has been implicated in experimental models of rheumatoid arthritis, experimental autoimmune encephalomyelitis (EAE), inflammatory bowel disease, asthma, and cardiovascular inflammation. In biomedical research, rat IL-17A serves as an important biomarker and mechanistic mediator of Th17-driven immune activation, neutrophilic inflammation, and autoimmune pathology in translational disease models.
