  • The Swine CCL3L1 (Macrophage Inflammatory Protein-1α, MIP-1α) ELISA is designed for the quantitative measurement of swine CCL3L1 in cell culture supernatants and can also be used to measure CCL3L1 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, providing flexibility for a wide range of immunology, inflammation, and translational research applications. CCL3L1 is a chemokine primarily produced by activated monocytes, macrophages, and T cells, and it functions as a potent mediator of leukocyte chemotaxis, particularly recruiting monocytes, T cells, and NK cells to sites of infection or tissue injury. In swine models, CCL3L1 is widely studied in the context of infectious disease, vaccine response, inflammatory disorders, and immune activation, where its expression reflects the intensity of innate and adaptive immune responses. Measurement of CCL3L1 provides valuable insight into leukocyte trafficking, inflammatory signaling, and immune system dynamics. Reported measurements of swine CCL3L1 have detected low to moderate picogram per milliliter concentrations in serum and plasma under basal conditions, with significantly elevated levels following immune stimulation, infection, or tissue inflammation, and measurable expression has also been observed in tissue lysates from inflamed or infected organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Swine CCL3L1 (C-C motif chemokine ligand 3-like 1, also known as LD78β or MIP-1αP) is a chemokine closely related to CCL3 (MIP-1α) that plays a role in leukocyte recruitment and regulation of inflammatory immune responses in pigs (Sus scrofa domesticus). CCL3L1 primarily signals through the CCR5 receptor, and to a lesser extent CCR1, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection or tissue injury. In swine, CCL3L1 is produced by activated macrophages, dendritic cells, and lymphocytes in response to inflammatory stimuli such as pathogen-associated molecular patterns and cytokines including TNF-α and IL-1β. CCL3L1-mediated signaling contributes to host defense by coordinating immune cell trafficking and amplifying inflammatory responses during infections, including porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus, and bacterial respiratory pathogens involved in porcine respiratory disease complex. Because CCR5-mediated chemokine signaling influences antiviral immunity and immune cell activation, CCL3L1 may also participate in regulation of inflammatory lung responses during respiratory infections. As a regulator of leukocyte migration and inflammatory signaling, swine CCL3L1 is relevant in studies of host–pathogen interactions, immune regulation, and vaccine responses, and contributes to understanding chemokine networks that influence disease resistance and immune function in pigs.
