  • The Swine CCL4 (Macrophage Inflammatory Protein-1β, MIP-1β) ELISA is designed for the quantitative measurement of swine CCL4 in cell culture supernatants and can also be used to measure CCL4 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, providing flexibility for a wide range of immunology, inflammation, and translational research applications. CCL4 is a chemokine primarily produced by activated monocytes, macrophages, and T cells, and it functions as a key mediator of leukocyte recruitment, particularly of monocytes, NK cells, and T lymphocytes, to sites of infection or tissue injury. In swine models, CCL4 is widely studied in the context of infectious disease, vaccine response, inflammatory disorders, and immune activation, where its expression reflects the magnitude and dynamics of innate and adaptive immune responses. Measurement of CCL4 provides valuable insight into leukocyte trafficking, inflammatory signaling, and immune system activation. Reported measurements of swine CCL4 have detected low to moderate picogram per milliliter concentrations in serum and plasma under basal conditions, with significantly elevated levels following immune stimulation, infection, or tissue inflammation, and measurable expression has also been observed in tissue lysates from inflamed or infected organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Swine CCL4 (C-C motif chemokine ligand 4, also known as MIP-1β, macrophage inflammatory protein-1 beta) is a pro-inflammatory chemokine that regulates recruitment and activation of immune cells during inflammatory and infectious responses in pigs (Sus scrofa domesticus). CCL4 primarily signals through the CCR5 receptor, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection, inflammation, or tissue injury. In pigs, CCL4 is produced by activated macrophages, dendritic cells, epithelial cells, and lymphocytes in response to inflammatory stimuli such as pathogen-associated molecules and cytokines including TNF-α and IL-1β. CCL4-mediated immune cell recruitment is particularly relevant in respiratory and systemic infectious diseases, including porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus, and bacterial pathogens involved in porcine respiratory disease complex, where immune cell trafficking to infected tissues contributes to antiviral and antibacterial defense. While CCL4-driven leukocyte migration supports pathogen clearance and immune activation, excessive or prolonged expression may contribute to inflammatory tissue damage, particularly in lung tissue during severe respiratory infections. Because pigs are widely used as large-animal models for human infectious and inflammatory diseases, characterization of swine CCL4 supports studies of chemokine-mediated immune cell migration, host–pathogen interactions, and development of vaccines and immunomodulatory therapies relevant to both veterinary medicine and translational biomedical research.
