  • The Swine IL-4 (Interleukin-4) ELISA is designed for the quantitative measurement of swine IL-4 in cell culture supernatants and can also be used to measure IL-4 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology, allergy, and translational research applications. IL-4 is a hallmark Th2 cytokine produced primarily by activated CD4⁺ T cells, mast cells, and basophils, and it plays a central role in Th2 differentiation, B cell proliferation, class switching to IgE, and macrophage polarization toward an alternatively activated (M2) phenotype. In swine models, IL-4 is widely studied in the context of allergic responses, parasitic infection, vaccine response, and immune regulation, where its expression reflects Th2-skewed immunity. Measurement of IL-4 provides valuable insight into immune polarization, humoral immune responses, and the balance between pro-inflammatory and regulatory pathways. Reported measurements of swine IL-4 have detected basal serum and plasma levels of approximately 10-80 pg/mL, with elevated levels-often 100-500 pg/mL-following immune stimulation, parasitic infection, or allergic challenge, and measurable IL-4 expression has also been observed in tissue lysates from inflamed or Th2-activated organs. Swine Interleukin-4 (IL-4) is a Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells, mast cells, and basophils in pigs (Sus scrofa domesticus), where it plays a central role in regulating humoral immunity, alternative macrophage activation, and modulation of inflammatory responses. IL-4 signals through receptor complexes containing IL-4Rα paired with the common gamma chain (γc) or IL-13Rα1, activating JAK/STAT6 pathways that promote B-cell proliferation, antibody production, and suppression of Th1-associated cytokines such as IFN-γ. In swine health, IL-4 is particularly important in responses to gastrointestinal and respiratory parasitic infections, where Th2 polarization supports parasite clearance and mucosal repair.
