  • The Swine IL-8 (Interleukin-8, CXCL8) ELISA is designed for the quantitative measurement of swine IL-8 in cell culture supernatants and can also be used to measure IL-8 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology, inflammation, and translational research applications. IL-8 is a potent pro-inflammatory chemokine primarily produced by activated monocytes, macrophages, epithelial cells, and endothelial cells, and it functions as a key mediator of neutrophil chemotaxis and activation, promoting the recruitment of neutrophils to sites of infection, tissue injury, or inflammation. In swine models, IL-8 is widely studied in the context of infectious disease, acute inflammation, immune activation, and tissue injury, where its expression reflects innate immune responses and inflammatory signaling. Measurement of IL-8 provides valuable insight into leukocyte trafficking, inflammation severity, and therapeutic intervention efficacy. Reported measurements of swine IL-8 have detected basal serum and plasma levels of approximately 50-300 pg/mL, with substantially elevated levels-often 1,000-5,000 pg/mL-following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or infected organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Swine Interleukin-8 (IL-8), also known as CXCL8, is a potent pro-inflammatory chemokine that plays a central role in innate immune responses in pigs (Sus scrofa domesticus) by mediating neutrophil chemotaxis, activation, and migration to sites of infection or tissue injury. Produced by macrophages, respiratory and intestinal epithelial cells, endothelial cells, and other immune cells in response to microbial products and inflammatory cytokines, swine IL-8 signals primarily through CXCR1 and CXCR2 receptors to promote rapid neutrophil recruitment and amplification of inflammatory responses. Elevated IL-8 expression is observed in economically significant diseases such as porcine reproductive and respiratory syndrome (PRRS), swine influenza, Mycoplasma hyopneumoniae infection, Actinobacillus pleuropneumoniae pneumonia, enteric infections, and bacterial sepsis, where excessive neutrophilic inflammation can contribute to lung injury, reduced growth performance, and systemic pathology. As a biomarker, IL-8 is widely used to assess immune activation, vaccine response, and disease severity in swine production systems. In translational research, pigs serve as valuable large-animal models for human respiratory disease, sepsis, trauma, and inflammatory disorders due to their physiological and immunological similarities to humans; thus, characterization of swine IL-8 supports studies of neutrophil-driven immunopathology, cytokine network regulation, and development of targeted anti-inflammatory and immunomodulatory therapies relevant to both veterinary and human medicine.
