  • The Swine IL-13 (Interleukin-13) ELISA is designed for the quantitative measurement of swine IL-13 in cell culture supernatants and can also be used to measure IL-13 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology, allergy, and translational research applications. IL-13 is a Th2 cytokine primarily produced by activated CD4⁺ T cells, mast cells, and basophils, and it plays a central role in B cell proliferation, class switching to IgE, mucus production, and alternatively activated macrophage (M2) polarization, contributing to allergic responses and humoral immunity. In swine models, IL-13 is widely studied in the context of allergic diseases, parasitic infections, asthma, and immune regulation, where its expression reflects Th2-skewed immune responses. Measurement of IL-13 provides valuable insight into immune polarization, allergic inflammation, and the balance between Th1 and Th2 immunity. Reported measurements of swine IL-13 have detected basal serum and plasma levels of approximately 20-150 pg/mL, with elevated levels-often 200-800 pg/mL-following immune stimulation, allergen exposure, or parasitic challenge, and measurable expression has also been observed in tissue lysates from Th2-activated or inflamed organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Swine Interleukin-13 (IL-13) is a Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells and other type 2 immune populations in pigs (Sus scrofa domesticus), where it plays an important role in regulating mucosal immunity, alternative macrophage activation, and tissue remodeling. IL-13 signals through receptor complexes composed of IL-13Rα1 and IL-4Rα, activating JAK/STAT6 pathways that promote IgE-associated responses, mucus production, modulation of pro-inflammatory cytokine signaling, and support of wound healing processes. In swine health, IL-13 is particularly relevant in responses to gastrointestinal and respiratory parasitic infections, where Th2 polarization aids parasite clearance and mucosal repair. IL-13 may also influence immune regulation during viral infections such as porcine reproductive and respiratory syndrome virus (PRRSV) and swine influenza by shaping the balance between Th1 and Th2 responses. While protective in controlling inflammation and supporting tissue repair, excessive or prolonged IL-13 activity may contribute to airway remodeling or fibrotic changes. As a biomarker of type 2 immune activation, swine IL-13 is important in studies of vaccine efficacy, host–parasite interactions, and immune resilience in production systems. In translational research, pigs serve as valuable large-animal models due to physiological similarities to humans, and characterization of swine IL-13 supports comparative investigations into Th2-mediated inflammation, fibrosis, and cytokine-targeted therapeutic strategies.
