  • The Swine IL-17A ELISA is designed for the quantitative measurement of swine IL-17A in cell culture supernatants and can also be used to measure IL-17A in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology, inflammation, and translational research applications. IL-17A is a signature cytokine of Th17 cells and plays a central role in pro-inflammatory immune responses, particularly at mucosal and barrier surfaces, by promoting the recruitment and activation of neutrophils and stimulating epithelial, endothelial, and stromal cells to produce chemokines, cytokines, and antimicrobial peptides. In swine models, IL-17A is widely studied in the context of autoimmune and inflammatory diseases, infectious disease, allergy, vaccine responses, and mucosal immunity, where dysregulated IL-17A signaling contributes to tissue inflammation, host defense, and immunopathology. Measurement of IL-17A provides critical insight into Th17 pathway activation, inflammatory immune dynamics, and immune regulation. Swine IL-17A shares 100% sequence homology with the common warthog, supporting precise cross-species comparison studies. Reported measurements of swine IL-17A have detected basal serum and plasma levels of approximately 10-80 pg/mL, with substantially elevated levels-often 200-1,000 pg/mL-following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or infected organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Swine IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays a critical role in host defense and inflammatory responses in pigs (Sus scrofa). IL-17A is produced primarily by Th17 CD4⁺ T cells, as well as by γδ T cells—which are abundant in swine—and other innate lymphoid cell populations in response to microbial infection and immune stimulation. It acts on epithelial, endothelial, and stromal cells to induce expression of pro-inflammatory cytokines (e.g., IL-6, TNF-α), chemokines (such as CXCL8), and antimicrobial peptides, thereby promoting neutrophil recruitment and enhancing mucosal barrier immunity in respiratory and gastrointestinal tissues. In healthy pigs, basal IL-17A expression is generally low, but levels increase during bacterial and fungal infections, enteric disease, respiratory inflammation, and parasitic challenge. IL-17A has been implicated in immune responses to pathogens such as porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus, Actinobacillus pleuropneumoniae, and enteric bacteria. In veterinary and translational research, swine IL-17A serves as an important biomarker of Th17-mediated immune activation and neutrophilic inflammation, providing insight into host–pathogen interactions, vaccine-induced immunity, and inflammatory disease mechanisms in both agricultural and biomedical research contexts.
