  • The Swine IL-22 (Interleukin-22) ELISA is designed for the quantitative measurement of swine IL-22 in cell culture supernatants and can also be used to measure IL-22 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology, mucosal immunity, and translational research applications. IL-22 is a cytokine primarily produced by Th17, Th22, and innate lymphoid cells, and it plays a central role in epithelial barrier integrity, tissue regeneration, antimicrobial defense, and inflammation at mucosal surfaces. In swine models, IL-22 is widely studied in the context of infectious disease, gut and respiratory inflammation, mucosal immunity, and tissue repair, where its expression reflects immune regulation at barrier surfaces. Measurement of IL-22 provides valuable insight into host defense mechanisms, epithelial cell function, and Th17/Th22 pathway activation. Swine IL-22 shares 100% sequence homology with the common warthog, supporting cross-species studies with high fidelity. Reported measurements of swine IL-22 have detected basal serum and plasma levels of approximately 20-150 pg/mL, with elevated levels-often 200-1,000 pg/mL-following immune stimulation, infection, or tissue inflammation, and measurable expression has also been observed in tissue lysates from mucosal and inflamed organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Swine IL-22 (Interleukin-22) is a member of the IL-10 cytokine family that plays a key role in mucosal immunity and epithelial barrier protection in pigs (Sus scrofa). IL-22 is produced primarily by activated CD4⁺ T cells, including Th17- and Th22-like subsets, as well as by γδ T cells and innate lymphoid cells (ILC3s), in response to microbial infection and inflammatory stimulation. It signals through the IL-22 receptor complex (IL-22R1 and IL-10R2), expressed mainly on epithelial and stromal cells, activating downstream JAK/STAT pathways—particularly STAT3—to induce antimicrobial peptide production, enhance chemokine expression, promote epithelial cell proliferation, and support tissue repair. In healthy swine, basal IL-22 expression is generally low but increases during bacterial, viral, and parasitic infections, particularly at mucosal surfaces of the gastrointestinal and respiratory tracts. Swine IL-22 has been implicated in immune responses to enteric pathogens such as Escherichia coli and Salmonella, as well as respiratory pathogens including porcine reproductive and respiratory syndrome virus (PRRSV) and swine influenza virus. In veterinary and translational research, swine IL-22 serves as a biomarker of mucosal immune activation and epithelial defense and is studied in the context of host–pathogen interactions, vaccine efficacy, and maintenance of barrier integrity in both agricultural and biomedical disease models.
