  • The Swine IL-10 (Interleukin-10) ELISA is designed for the quantitative measurement of swine IL-10 in cell culture supernatants and can also be used to measure IL-10 in additional biological sample types reported in the literature, including serum, plasma, tissue lysates, and bronchoalveolar lavage fluid, supporting a wide range of immunology, inflammation, and translational research applications. IL-10 is an anti-inflammatory cytokine primarily produced by regulatory T cells, monocytes, macrophages, and B cells, and it plays a central role in limiting immune responses, controlling inflammation, and maintaining immune homeostasis. In swine models, IL-10 is widely studied in the context of infectious disease, vaccine responses, autoimmune disorders, sepsis, and inflammatory conditions, where its expression reflects regulatory immune activity and the balance between pro- and anti-inflammatory pathways. Measurement of IL-10 provides valuable insight into immune regulation, disease progression, and the effectiveness of immunomodulatory interventions. Reported measurements of swine IL-10 have detected basal serum and plasma levels of approximately 10-100 pg/mL, with elevated levels-often 200-1,000 pg/mL-following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or infected organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Swine Interleukin-10 (IL-10) is a key anti-inflammatory cytokine produced by regulatory T cells (Tregs), Th2 cells, macrophages, dendritic cells, and certain B-cell populations in pigs (Sus scrofa domesticus), where it plays a central role in limiting excessive immune activation and maintaining immune homeostasis. IL-10 signals through the IL-10 receptor complex (IL-10R1/IL-10R2), activating JAK/STAT3 pathways that suppress pro-inflammatory cytokine production (including IL-1β, TNF-α, IL-6, and IFN-γ), reduce antigen presentation, and downregulate Th1-type cell-mediated responses. In swine health, IL-10 is critically involved in modulating immune responses during economically significant infections such as porcine reproductive and respiratory syndrome virus (PRRSV), African swine fever virus (ASFV), swine influenza virus, enteric infections, and bacterial pneumonia, where elevated IL-10 can limit tissue damage but may also contribute to immune suppression and viral persistence. The balance between IL-10 and pro-inflammatory cytokines influences disease severity, vaccine responsiveness, and growth performance. As a biomarker of immune regulation, swine IL-10 is widely used in studies of vaccine efficacy, immunopathogenesis, and herd health management.
