  • The Canine IL-1RA (Interleukin-1 Receptor Antagonist) ELISA is designed for the quantitative measurement of canine IL-1RA in cell culture supernatants and can also be used to measure IL-1RA in additional biological sample types reported in the literature, including serum, plasma, and tissue lysates, supporting a wide range of immunology, inflammation, and translational research applications. IL-1RA is an anti-inflammatory cytokine produced primarily by monocytes, macrophages, and fibroblasts, and it functions as a natural inhibitor of IL-1α and IL-1β by competitively binding to IL-1 receptors without inducing signal transduction, thereby modulating immune responses and limiting excessive inflammation. In canine models, IL-1RA is widely studied in the context of autoimmune disease, sepsis, inflammatory conditions, and vaccine response, where its expression reflects the balance between pro- and anti-inflammatory signaling. Measurement of IL-1RA provides valuable insight into immune regulation, disease progression, and therapeutic efficacy, particularly in conditions characterized by excessive IL-1 activity. Canine IL-1RA shares 100% sequence homology with Canis lupus dingo (dingo), Nyctereutes procyonoides (raccoon dog), Vulpes lagopus (Arctic fox), and Vulpes vulpes (red fox), supporting precise cross-species studies. Reported measurements of canine IL-1RA have detected basal serum and plasma levels of approximately 50-400 pg/mL, with elevated levels-often 500-2,000 pg/mL-following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Canine Interleukin-1 Receptor Antagonist (IL-1Ra) is an anti-inflammatory cytokine produced by monocytes, macrophages, neutrophils, and other immune cells in dogs (Canis lupus familiaris) that serves as a natural regulator of IL-1–mediated inflammation. IL-1Ra competitively binds to the IL-1 receptor type 1 (IL-1R1) without activating downstream signaling, thereby blocking the pro-inflammatory effects of IL-1α and IL-1β and limiting activation of NF-κB and MAPK pathways that drive fever, leukocyte recruitment, and tissue inflammation. In canine health, IL-1Ra plays an important role in modulating inflammatory conditions such as osteoarthritis, immune-mediated polyarthritis, atopic dermatitis, inflammatory bowel disease, sepsis, and other inflammatory or infectious diseases where excessive IL-1 signaling contributes to pathology.
