  • The Equine IFN-α1 (Interferon Alpha 1) ELISA is designed for the quantitative measurement of equine IFN-α1 in cell culture supernatants and can also be used to measure IFN-α1 in additional biological sample types reported in the literature, including serum, plasma, and tissue lysates, supporting a wide range of immunology, antiviral, and translational research applications. IFN-α1 is a type I interferon primarily produced by plasmacytoid dendritic cells and other leukocytes, and it plays a central role in antiviral defense, innate immunity, and the activation of adaptive immune responses. In equine models, IFN-α1 is widely studied in the context of viral infections, vaccine responses, and immunotherapy, where its expression reflects the activation of antiviral signaling pathways and the host immune response. Measurement of IFN-α1 provides valuable insight into innate immune activation, viral pathogenesis, and the efficacy of immunomodulatory interventions. Reported measurements of equine IFN-α1 have detected basal serum and plasma levels of approximately 20-200 pg/mL, with elevated levels-often 300-2,500 pg/mL-following viral infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from infected or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Equine Interferon-Alpha (IFN-α) is a type I interferon produced primarily by plasmacytoid dendritic cells and virus-infected epithelial and immune cells in horses (Equus caballus), where it plays a central role in early innate antiviral defense. Upon binding to the type I interferon receptor complex (IFNAR1/IFNAR2), equine IFN-α activates JAK/STAT signaling pathways, leading to induction of interferon-stimulated genes (ISGs) that inhibit viral replication, enhance antigen presentation, and promote activation of natural killer (NK) cells and adaptive immune responses. IFN-α is rapidly upregulated in response to important equine viral pathogens such as equine influenza virus, equine herpesvirus-1 and -4 (EHV-1/EHV-4), West Nile virus, and equine arteritis virus, where it contributes to viral control but may also influence inflammatory pathology if dysregulated. Modulation of type I interferon responses can affect disease severity, viral shedding, and vaccine responsiveness. In veterinary and translational research, characterization of equine IFN-α supports studies of respiratory viral pathogenesis, neuroinvasive viral disease (e.g., EHV-1 and West Nile virus), antiviral therapeutic development, and comparative type I interferon biology in large-animal systems relevant to both equine health and broader immunological research.
