  • The Canine CCL4 (Chemokine (C-C motif) Ligand 4, also known as MIP-1β) ELISA is designed for the quantitative measurement of canine CCL4 in cell culture supernatants and can also be used to measure CCL4 in additional biological sample types reported in the literature, including serum, plasma, and tissue lysates, supporting a wide range of immunology, inflammation, and translational research applications. CCL4 is a chemokine primarily produced by activated T cells, natural killer cells, monocytes, and macrophages, and it plays a central role in leukocyte chemotaxis, immune cell activation, and inflammatory signaling. In canine models, CCL4 is widely studied in the context of viral and bacterial infections, autoimmune disorders, and inflammatory diseases, where its expression reflects immune cell recruitment and inflammatory response dynamics. Measurement of CCL4 provides valuable insight into immune activation, disease progression, and the efficacy of immunomodulatory interventions. Canine CCL4 shares 100% sequence homology with Canis lupus dingo (dingo), Vulpes lagopus (Arctic fox), and Vulpes vulpes (red fox), which is important for cross-species studies, comparative immunology, and translational research because assays developed for domestic dogs can reliably detect and quantify CCL4 in these related species. Reported measurements of canine CCL4 have detected basal serum and plasma levels of approximately 50-300 pg/mL, with elevated levels-often 500-2,500 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Canine CCL4 (C-C motif chemokine ligand 4, also known as MIP-1β, macrophage inflammatory protein-1 beta) is a pro-inflammatory chemokine that plays an important role in recruitment and activation of immune cells during inflammatory and antiviral immune responses in dogs (Canis lupus familiaris). CCL4 primarily signals through the CCR5 receptor, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection or tissue injury. In dogs, CCL4 is produced by activated macrophages, dendritic cells, epithelial cells, and lymphocytes in response to inflammatory stimuli such as microbial components and cytokines including TNF-α and IL-1β. CCL4-mediated leukocyte recruitment contributes to host defense in infectious diseases such as canine distemper virus, parvovirus infection, and bacterial infections, where coordinated immune cell trafficking supports pathogen clearance. CCL4 is also implicated in chronic inflammatory and immune-mediated conditions, including dermatitis and inflammatory bowel disease, as well as in canine cancers such as lymphoma, where chemokine signaling can influence immune cell infiltration within the tumor microenvironment. While CCL4-driven leukocyte migration is important for immune activation and pathogen defense, excessive or prolonged expression may contribute to tissue inflammation and damage. As a regulator of immune cell trafficking and inflammatory signaling, canine CCL4 is important for studies of host–pathogen interactions, immune regulation, and inflammatory disease mechanisms relevant to both veterinary medicine and comparative immunology.
