  • The Rat CCL4 (Chemokine (C-C motif) Ligand 4, also known as MIP-1β) ELISA is designed for the quantitative measurement of rat CCL4 in cell culture supernatants and can also be used to measure CCL4 in additional biological sample types reported in the literature, including serum, plasma, and tissue lysates, supporting a wide range of immunology, inflammation, and translational research applications. CCL4 is a chemokine primarily produced by activated T cells, natural killer cells, monocytes, and macrophages, and it plays a central role in leukocyte chemotaxis, immune cell activation, and inflammatory signaling. In rat models, CCL4 is widely studied in the context of viral and bacterial infections, autoimmune disorders, and inflammatory diseases, where its expression reflects immune cell recruitment and inflammatory response dynamics. Measurement of CCL4 provides valuable insight into immune activation, disease progression, and the efficacy of immunomodulatory interventions. Reported measurements of rat CCL4 have detected basal serum and plasma levels of approximately 20-200 pg/mL, with elevated levels-often 300-2,000 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Rat CCL4 (C-C motif chemokine ligand 4, also known as MIP-1β, macrophage inflammatory protein-1 beta) is a pro-inflammatory chemokine that regulates recruitment and activation of immune cells during inflammatory and infectious responses in rats (Rattus norvegicus). CCL4 primarily signals through the CCR5 receptor, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection, inflammation, or tissue injury. In rats, CCL4 is produced by activated macrophages, dendritic cells, T lymphocytes, epithelial cells, and endothelial cells in response to inflammatory stimuli such as microbial products and cytokines including TNF-α and IL-1β. Rat CCL4 is widely studied in experimental models of neuroinflammation, autoimmune disease, cardiovascular injury, and infectious disease, where chemokine-driven immune cell recruitment contributes to inflammatory signaling and tissue pathology. It has been implicated in models of neuropathic pain, stroke, and central nervous system injury, where CCL4-mediated leukocyte trafficking influences neuroimmune interactions and inflammatory responses in the brain and spinal cord. While CCL4-driven immune cell migration supports host defense and tissue repair, excessive or prolonged expression may contribute to chronic inflammation and tissue damage. Because rats are widely used in neuroscience, pharmacology, and inflammatory disease research, characterization of rat CCL4 supports investigations into CCR5-mediated immune cell trafficking, inflammatory signaling pathways, and host–pathogen interactions relevant to human disease.
