  • The Ovine CXCL10 (C-X-C Motif Chemokine Ligand 10, also known as IP-10) ELISA is designed for the quantitative measurement of CXCL10 in sheep cell culture supernatants and can also be used to measure CXCL10 in additional biological sample types reported in the literature, including serum, plasma, and tissue lysates, supporting a wide range of immunology, inflammation, and translational research applications. CXCL10 is a chemokine primarily produced by monocytes, macrophages, endothelial cells, and fibroblasts in response to interferon-γ stimulation, and it plays a central role in chemoattraction of activated T cells, NK cells, and monocytes, as well as regulation of immune responses during infection and inflammation. In sheep models, CXCL10 is widely studied in the context of viral and bacterial infections, autoimmune disorders, and inflammatory diseases, where its expression reflects immune cell recruitment and inflammatory signaling. Measurement of CXCL10 provides valuable insight into immune activation, disease progression, and the efficacy of immunomodulatory interventions. Reported measurements of ovine CXCL10 have detected basal serum and plasma levels of approximately 50-400 pg/mL, with elevated levels-often 500-2,500 pg/mL-following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements.   • Ovine CXCL10 (C-X-C motif chemokine ligand 10), also known as IP-10 (Interferon gamma-induced protein 10), is a proinflammatory chemokine belonging to the CXC chemokine family, which also includes CXCL9 (MIG) and CXCL11 (I-TAC) that regulate T-cell recruitment during antiviral and inflammatory immune responses. In sheep (Ovis aries), CXCL10 is produced by macrophages, dendritic cells, epithelial cells, endothelial cells, and fibroblasts in response to interferon-γ (IFN-γ), microbial infection, or inflammatory cytokines. CXCL10 signals primarily through the chemokine receptor CXCR3, which is expressed on activated T lymphocytes, natural killer (NK) cells, and other immune cells, promoting chemotaxis and activation of these cells at sites of infection or inflammation. In ovine health, CXCL10 contributes to immune responses against viral, bacterial, and parasitic pathogens, including infections affecting the respiratory tract and gastrointestinal system. Sheep are also used as large-animal models for inflammatory lung diseases, and CXCL10 expression has been examined in ovine models of respiratory syncytial virus (RSV) infection, which mimic aspects of human infant RSV disease, where CXCL10-mediated recruitment of CXCR3⁺ T cells contributes to antiviral immunity and airway inflammation. These studies make ovine CXCL10 relevant for comparative immunology, respiratory disease research, and investigation of interferon-driven immune responses.
