  • The Canine IL-1α (Interleukin-1 Alpha) ELISA is designed for the quantitative measurement of IL-1α in dog cell culture supernatants and can also be used to measure IL-1α in additional biological sample types reported in the literature, including serum, plasma, and tissue lysates, supporting a wide range of immunology, inflammation, and translational research applications. IL-1α is a pro-inflammatory cytokine primarily produced by activated monocytes, macrophages, and epithelial cells, and it plays a central role in initiating and amplifying inflammatory responses, recruiting immune cells, and regulating cytokine networks. In the canine model, IL-1α is widely studied in the context of infectious diseases, autoimmune disorders, cancer, and tissue injury, where its expression reflects the early stages of immune activation and local inflammation. Measurement of IL-1α provides valuable insight into disease progression, immune regulation, and the efficacy of anti-inflammatory or immunomodulatory therapies. Reported measurements of canine IL-1α have detected basal serum and plasma levels of approximately 20-150 pg/mL, with elevated levels-often 200-1,500 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Canine Interleukin-1 Alpha (IL-1α) is a pro-inflammatory cytokine belonging to the IL-1 family, which includes IL-1α, IL-1β, and the IL-1 receptor antagonist (IL-1Ra), and plays a key role in initiating and amplifying innate immune responses in dogs (Canis lupus familiaris). IL-1α is constitutively expressed in epithelial and barrier tissues and functions as an intracellular alarmin that is rapidly released upon cell injury or necrosis, triggering early inflammatory signaling. It binds to the IL-1 receptor type 1 (IL-1R1), activating NF-κB and MAPK pathways that promote leukocyte recruitment, endothelial activation, fever, and production of additional pro-inflammatory mediators. In canine health, IL-1α is particularly relevant in inflammatory skin diseases such as atopic dermatitis, wound healing responses, osteoarthritis, inflammatory bowel disease, and certain neoplastic conditions where tumor-associated inflammation contributes to disease progression. While essential for pathogen defense and tissue repair, excessive or chronic IL-1α signaling can exacerbate tissue damage and perpetuate inflammation. As an early mediator of sterile and infectious inflammation, canine IL-1α serves as a biomarker of tissue injury and innate immune activation. In translational research, dogs are valuable spontaneous models for human dermatologic disease, osteoarthritis, inflammatory bowel disease, and cancer, and characterization of canine IL-1α supports comparative studies of cytokine-driven inflammation and development of targeted anti-inflammatory therapies relevant to both veterinary and human medicine.
