  • The Mouse IL-1β (Interleukin-1 Beta) ELISA is designed for the quantitative measurement of IL-1β in cell culture supernatants from mice and can also be used to measure IL-1β in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, cerebrospinal fluid, synovial fluid, bronchoalveolar lavage fluid, and tissue lysates from organs such as liver, spleen, lung, kidney, and lymph nodes, supporting a broad range of immunology, inflammation, and translational research applications. IL-1β is a pro-inflammatory cytokine primarily produced by activated monocytes, macrophages, and dendritic cells, and it plays a central role in initiating and amplifying inflammatory responses, activating immune cells, and regulating cytokine networks. In the mouse model, IL-1β is widely studied in the context of infectious diseases, autoimmune disorders, metabolic inflammation, and tissue injury, where its expression reflects early immune activation and inflammatory signaling. Measurement of IL-1β provides valuable insight into disease progression, immune regulation, and the efficacy of anti-inflammatory or immunomodulatory therapies. Reported measurements of mouse IL-1β have detected basal serum and plasma levels of approximately 10-150 pg/mL, with elevated levels-often 200-1,500 pg/mL-following infection, immune stimulation, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Mouse Interleukin-1 Beta (IL-1β) is a potent pro-inflammatory cytokine produced primarily by activated macrophages, monocytes, dendritic cells, and microglia in mice (Mus musculus) in response to infection, tissue damage, or danger-associated molecular patterns. Synthesized as an inactive precursor (pro–IL-1β), it is cleaved into its active form by caspase-1 within inflammasome complexes—most notably the NLRP3 inflammasome—and signals through the IL-1 receptor type 1 (IL-1R1), activating NF-κB and MAPK pathways that promote fever, leukocyte recruitment, endothelial activation, and amplification of inflammatory responses. Murine IL-1β is central to widely used experimental models of sepsis, gout, rheumatoid arthritis, inflammatory bowel disease, neuroinflammation, metabolic disorders, and cancer, where dysregulated IL-1β production contributes to tissue injury and chronic inflammation. IL-1β–deficient, IL-1R1 knockout, and inflammasome-deficient mouse models have been instrumental in defining the molecular mechanisms of inflammasome activation and cytokine-driven pathology. As both a biomarker and therapeutic target, mouse IL-1β remains foundational in preclinical research investigating anti-inflammatory drugs, biologics targeting IL-1 signaling, and the broader role of innate immune activation in infectious and inflammatory diseases relevant to human health.
