  • The Equine CCL5 (Chemokine (C-C motif) Ligand 5, also known as RANTES) ELISA is designed for the quantitative measurement of CCL5 in cell culture supernatants from horses and can also be used to measure CCL5 in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, synovial fluid, bronchoalveolar lavage fluid, and tissue lysates from organs such as lung, spleen, liver, and lymph nodes, supporting a broad range of immunology, inflammation, and translational research applications. CCL5 is a chemokine primarily produced by T cells, macrophages, platelets, and endothelial cells, and it plays a central role in recruiting T cells, eosinophils, and basophils to sites of inflammation, as well as modulating immune responses during infection and tissue injury. In the equine model, CCL5 is widely studied in the context of viral and bacterial infections, allergic responses, and autoimmune disorders, where its expression reflects immune activation and inflammatory signaling. Measurement of CCL5 provides valuable insight into immune regulation, disease progression, and the efficacy of immunomodulatory therapies. Reported measurements of equine CCL5 have detected basal serum and plasma levels of approximately 50-400 pg/mL, with elevated levels-often 500-2,500 pg/mL-following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from inflamed or immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to optimize assay sensitivity and dynamic range based on their specific sample type and experimental requirements. Equine CCL5 (C–C motif chemokine ligand 5, also known as RANTES – Regulated upon Activation, Normal T cell Expressed and Secreted) is a proinflammatory chemokine that regulates immune cell recruitment and inflammatory responses in horses (Equus caballus). CCL5 is a member of the CC chemokine family, which also includes CCL3 (MIP-1α), CCL4 (MIP-1β), and CCL2 (MCP-1), chemokines that coordinate leukocyte trafficking during immune activation. In horses, CCL5 is produced primarily by activated T lymphocytes, macrophages, dendritic cells, platelets, and epithelial cells in response to infection, inflammatory stimuli, or tissue injury, and it exerts its biological effects mainly through binding to the chemokine receptors CCR1, CCR3, and CCR5, promoting chemotaxis and activation of T cells, monocytes/macrophages, eosinophils, and natural killer (NK) cells at sites of inflammation. In equine health, CCL5 contributes to immune responses associated with respiratory diseases such as equine asthma (recurrent airway obstruction), viral infections, and inflammatory airway conditions, where chemokine-mediated recruitment of leukocytes to lung tissues is a key component of the inflammatory response. Because horses are commonly studied in respiratory disease, inflammatory disorders, and comparative immunology, equine CCL5 is investigated in research on chemokine-regulated leukocyte trafficking, airway inflammation, and immune responses to infectious and environmental triggers.
