  • The Dolphin IL-2 (Interleukin-2) ELISA is designed for the quantitative measurement of IL-2 in cell culture supernatants from dolphins and can also be used to measure IL-2 in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, liver, and lung, supporting a broad range of immunology, inflammation, and marine mammal translational research applications. IL-2 is a cytokine primarily produced by activated CD4⁺ T cells, and to a lesser extent by CD8⁺ T cells, and it plays a central role in stimulating T cell proliferation, promoting regulatory T cell function, and supporting adaptive immune responses. In the dolphin animal model, IL-2 is widely studied in the context of immune responses to viral and bacterial infections, vaccination, and immunomodulatory therapies, where its expression reflects immune system activation and adaptive immune regulation. Measurement of IL-2 provides valuable insight into immune competence, disease progression, and the efficacy of vaccines or immunotherapies in dolphins. Reported measurements of dolphin IL-2 have detected basal serum and plasma levels of approximately 5-100 pg/mL, with elevated levels-often 150-1,200 pg/mL-following immune stimulation, infection, or inflammatory challenge, and measurable expression has also been observed in tissue lysates from immune-activated organs. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-2 in other sample types to ensure optimal sensitivity and accuracy. Optimal buffers, antibody concentrations, incubation times, temperatures, and assay conditions have not been predetermined, allowing researchers to further adjust assay parameters based on their specific sample type and experimental requirements. Dolphin IL-2 (Interleukin-2) is a key T cell–derived cytokine that regulates lymphocyte proliferation, differentiation, and immune homeostasis in dolphins (e.g., Tursiops truncatus). IL-2 is produced primarily by activated CD4⁺ and CD8⁺ T lymphocytes following antigen recognition and costimulatory signaling, and it functions through the IL-2 receptor complex (IL-2Rα/CD25, IL-2Rβ, and the common γ chain) to promote clonal expansion of antigen-specific T cells, enhance cytotoxic T lymphocyte and natural killer (NK) cell activity, and support development and maintenance of regulatory T cells (Tregs). In healthy dolphins, basal IL-2 expression is expected to be low but increases during infection, inflammatory stimulation, or environmental stress exposure. Altered IL-2 responses may occur during viral or bacterial infections, chronic inflammatory conditions, or immune challenges observed in both managed and free-ranging populations. Dolphins serve as valuable comparative models for studying marine mammal immunology, environmental immunotoxicology, and host responses to emerging infectious diseases. In marine mammal and translational research, dolphin IL-2 is used as a biomarker of T cell activation and cell-mediated immunity, providing insight into adaptive immune function and immune system health in marine species.
