  • The Human IFN-γ (Interferon gamma) ELISA is designed for the quantitative measurement of IFN-γ in cell culture supernatants from human cells and can also be used to measure IFN-γ in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, peripheral blood mononuclear cell (PBMC) culture supernatants, bronchoalveolar lavage fluid, and tissue lysates. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IFN-γ in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IFN-γ is a key immunoregulatory cytokine produced primarily by activated T lymphocytes, natural killer (NK) cells, and cytotoxic T cells, and it plays a central role in Th1-mediated immune responses, macrophage activation, antigen presentation, and host defense against intracellular pathogens. IFN-γ is widely studied in the context of infectious disease, cancer immunology, autoimmune and inflammatory disorders, vaccine development, and immunotherapy, where it serves as a critical biomarker of cell-mediated immune activation. Measurement of IFN-γ provides valuable insight into immune status, cytokine signaling dynamics, disease progression, and therapeutic efficacy, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing to tens to hundreds of picograms per milliliter following immune stimulation or during active inflammatory or infectious states, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Human Interferon-γ (IFN-γ) is a type II interferon cytokine that plays a central role in cell-mediated immunity and regulation of antimicrobial and inflammatory responses in humans (Homo sapiens). IFN-γ is produced primarily by activated T helper 1 (Th1) cells, cytotoxic T lymphocytes, and natural killer (NK) cells following stimulation by intracellular pathogens or immune signaling molecules such as IL-12 and IL-18. Human IFN-γ binds to the interferon-γ receptor (IFNGR1/IFNGR2) expressed on many immune and non-immune cells, activating intracellular signaling through the JAK–STAT pathway, particularly STAT1, which induces expression of numerous interferon-stimulated genes involved in antigen presentation, macrophage activation, and antimicrobial defense. IFN-γ is critical for activation of macrophages, enhancement of MHC class I and II expression, and control of intracellular pathogens, especially mycobacteria, viruses, and certain parasites. In human health, IFN-γ is essential for immunity against diseases such as tuberculosis and viral infections, and defects in IFN-γ signaling can lead to increased susceptibility to mycobacterial disease. Human IFN-γ is also widely studied and used in clinical diagnostics and immunotherapy, including IFN-γ release assays (IGRAs) for detection of Mycobacterium tuberculosis infection and therapeutic use of recombinant IFN-γ in certain immune deficiency and infectious disease conditions.
