  • The Dolphin CXCL10 (C-X-C motif chemokine ligand 10, also known as IP-10) ELISA is designed for the quantitative measurement of CXCL10 in cell culture supernatants from dolphins and can also be used to measure CXCL10 in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, liver, lung, and lymph nodes. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CXCL10 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. CXCL10 is a chemokine primarily produced by monocytes, macrophages, endothelial cells, and fibroblasts in response to interferon signaling, and it plays a central role in recruiting activated T cells, natural killer (NK) cells, and other immune cells to sites of inflammation or infection. In the dolphin animal model, CXCL10 is widely studied in the context of viral and bacterial infections, inflammatory responses, and immune regulation, where its expression reflects the activation of interferon-mediated signaling pathways. Measurement of CXCL10 provides valuable insight into immune activation, inflammatory status, disease progression, and the efficacy of immunomodulatory or antiviral interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing to tens to hundreds of picograms per milliliter in serum, plasma, or tissue-derived samples during infection or inflammatory challenge. Dolphin CXCL10 (C-X-C motif chemokine ligand 10), also known as IP-10 (Interferon gamma–induced protein 10), is a proinflammatory chemokine belonging to the CXC chemokine family, which also includes CXCL9 (MIG) and CXCL11 (I-TAC) that regulate T-cell recruitment during antiviral and inflammatory immune responses. In dolphins, particularly the bottlenose dolphin (Tursiops truncatus), CXCL10 is produced by macrophages, dendritic cells, epithelial cells, endothelial cells, and fibroblasts in response to interferon-γ (IFN-γ), viral infection, or inflammatory cytokines. CXCL10 signals primarily through the chemokine receptor CXCR3, which is expressed on activated T lymphocytes, natural killer (NK) cells, and other immune cells, promoting chemotaxis and activation of these cells at sites of infection or inflammation. In dolphin health, CXCL10 likely contributes to immune responses against viral, bacterial, and parasitic pathogens encountered in marine environments.
