  • The Feline CD40 Ligand (CD40L, also known as CD154) ELISA is designed for the quantitative measurement of CD40L in cell culture supernatants from cats and can also be used to measure CD40L in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, and thymus. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CD40L in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. CD40L is a transmembrane protein primarily expressed by activated feline T lymphocytes and plays a central role in B cell activation, immunoglobulin class switching, dendritic cell maturation, and the regulation of adaptive immune responses. In the feline animal model, CD40L is widely studied in the context of immune responses to infection, vaccination, inflammatory conditions, and immunomodulatory therapies, where its expression reflects T cell activation and immune system modulation. The feline CD40L protein is 100% homologous to CD40L from Panthera pardus (leopard), Panthera tigris altaica (Amur tiger), Acinonyx jubatus (cheetah), Lynx canadensis (Canada lynx), and Lynx pardinus (Spanish lynx), supporting its use in comparative and cross-species studies across wild and domestic felids and enabling reliable extrapolation of immune response findings in the feline animal model. Measurement of feline CD40L provides valuable insight into immune activation, cytokine signaling, adaptive immune regulation, and the efficacy of immunotherapeutic or vaccine interventions, with reported levels in cats typically detected at low picogram-per-milliliter concentrations under basal conditions (~20-180 pg/mL in serum or plasma) and increasing to several hundred to over a thousand picograms per milliliter following immune stimulation or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant feline CD40L protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Feline CD40 Ligand (CD40L), also known as CD154 and encoded by the CD40LG gene, is a type II transmembrane protein belonging to the tumor necrosis factor (TNF) superfamily, which includes related ligands such as TNF-α, CD27 ligand (CD70), OX40 ligand (CD252), and 4-1BB ligand (CD137L) that regulate immune activation and cell–cell signaling. In cats (Felis catus), CD40L is primarily expressed on activated CD4⁺ T lymphocytes and can also be found on platelets and other immune cells following activation. CD40L binds to its receptor CD40, which is expressed on B cells, dendritic cells, macrophages, and other antigen-presenting cells, triggering intracellular signaling pathways such as NF-κB and MAPK that promote B-cell activation, immunoglobulin class switching, germinal center formation, and development of memory B cells. Through these interactions, feline CD40L plays a critical role in T cell–dependent antibody responses and coordination of adaptive immunity. In feline health, the CD40–CD40L signaling pathway is important for immune responses to viral infections such as feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV) as well as bacterial and parasitic pathogens. Because CD40L is essential for T-cell–mediated B-cell help and immune regulation, feline CD40L is studied in veterinary immunology, infectious disease research, and vaccine development, helping to improve understanding of adaptive immune responses and immunomodulatory signaling in cats.
