  • The Swine CD40 Ligand (CD40L, also known as CD154) ELISA is designed for the quantitative measurement of CD40L in cell culture supernatants from swine and can also be used to measure CD40L in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, and thymus. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CD40L in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. CD40L is a transmembrane protein primarily expressed by activated swine T lymphocytes and plays a central role in B cell activation, immunoglobulin class switching, dendritic cell maturation, and the regulation of adaptive immune responses. In the swine animal model, CD40L is widely studied in the context of immune responses to infection, vaccination, inflammatory conditions, and immunomodulatory therapies, where its expression reflects T cell activation and immune system modulation. Measurement of swine CD40L provides valuable insight into immune activation, cytokine signaling, adaptive immune regulation, and the efficacy of immunotherapeutic or vaccine interventions, with reported levels in pigs typically detected at low picogram-per-milliliter concentrations under basal conditions (~20-150 pg/mL in serum or plasma) and increasing to several hundred to over a thousand picograms per milliliter following immune stimulation or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant swine CD40L protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Swine CD40 Ligand (CD40L), also known as CD154 and encoded by the CD40LG gene, is a type II transmembrane protein belonging to the tumor necrosis factor (TNF) superfamily, which includes related ligands such as TNF-α, CD27 ligand (CD70), OX40 ligand (CD252), and 4-1BB ligand (CD137L) that regulate immune activation and cell–cell signaling. In pigs (Sus scrofa), CD40L is primarily expressed on activated CD4⁺ T lymphocytes, and can also be detected on platelets and other activated immune cells following antigen stimulation. CD40L binds to its receptor CD40, which is expressed on B cells, dendritic cells, macrophages, and other antigen-presenting cells, triggering intracellular signaling pathways such as NF-κB and MAPK that promote B-cell activation, proliferation, immunoglobulin class switching, germinal center formation, and the development of memory B cells. Through these mechanisms, swine CD40L plays a key role in T cell–dependent antibody responses and regulation of adaptive immunity. In swine health, the CD40–CD40L signaling pathway contributes to immune responses against viral and bacterial pathogens, including porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus, and other infectious agents affecting pigs. Because pigs share many physiological and immunological similarities with humans, swine CD40L is also studied in comparative immunology, vaccine development, and translational animal models, helping to improve understanding of costimulatory signaling and immune regulation in both veterinary and human medicine.
