  • The Equine IL-12p40 (Interleukin-12 p40 subunit) ELISA is designed for the quantitative measurement of IL-12p40 in cell culture supernatants from horses and can also be used to measure IL-12p40 in a wide variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, and liver. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-12p40 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-12p40 is a subunit shared by IL-12 and IL-23, produced primarily by activated monocytes, macrophages, and dendritic cells, and plays a central role in regulating Th1 immune responses, promoting interferon-gamma production, and directing adaptive immune activation. In the equine animal model, IL-12p40 is widely studied in the context of infectious disease, vaccine development, inflammatory disorders, and immunomodulatory therapies, where its measurement reflects cytokine signaling and immune system activation. Measurement of equine IL-12p40 provides valuable insight into immune regulation, cytokine network dynamics, disease progression, and the efficacy of immunotherapeutic or vaccine interventions, with reported levels in horses typically detected at low picogram-per-milliliter concentrations under basal conditions (~30-150 pg/mL in serum or plasma) and increasing to hundreds of picograms per milliliter following immune stimulation or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant equine IL-12p40 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Equine Interleukin-12/Interleukin-23 p40 (IL-12/IL-23 p40) is a cytokine subunit that plays an important role in regulating immune responses in horses (Equus caballus). The p40 subunit is shared by two heterodimeric cytokines, IL-12 and IL-23, where it pairs with the p35 subunit to form IL-12 and with the p19 subunit to form IL-23. These cytokines are produced primarily by activated antigen-presenting cells, including macrophages and dendritic cells, in response to microbial infection or inflammatory stimuli. Equine IL-12/IL-23 p40 contributes to activation of natural killer (NK) cells and differentiation of T helper cells toward Th1-type immune responses, promoting production of interferon-γ (IFN-γ) and enhancing cell-mediated immunity against intracellular pathogens. Through IL-23 signaling, the p40 subunit also supports maintenance and expansion of Th17 cells involved in inflammatory and host defense pathways. In horses, IL-12/IL-23 p40 participates in immune responses against bacterial, viral, and parasitic infections and is studied in the context of equine diseases such as Rhodococcus equi pneumonia and equine herpesvirus infections. Due to its role in regulating cellular immunity, equine IL-12/IL-23 p40 is widely studied in veterinary immunology and translational research, including investigations of host–pathogen interactions, vaccine responses, and immune-mediated inflammatory conditions relevant to both equine health and comparative biomedical research.
