  • The Zebrafish IFN-γ1-1 (Interferon gamma 1-1) ELISA is designed for the quantitative measurement of IFN-γ1-1 in cell culture supernatants from zebrafish (Danio rerio) and can also be used to measure IFN-γ1-1 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, kidney, gill, and liver. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IFN-γ1-1 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IFN-γ1-1 is a key immunoregulatory cytokine produced primarily by activated T lymphocytes and natural killer-like cells in zebrafish and plays a central role in innate and adaptive immune responses, pathogen defense, and cytokine-mediated signaling. In the zebrafish animal model, IFN-γ1-1 is widely studied in the context of infectious disease, immunotoxicology, vaccine response, and immune system development, where its expression reflects cellular immune activation and host defense mechanisms. Measurement of IFN-γ1-1 provides valuable insight into immune regulation, cytokine signaling, disease progression, and the efficacy of immunomodulatory interventions, with reported levels in zebrafish typically detected at low nanogram-per-milliliter concentrations under basal conditions (~0.1-1 ng/mL in serum or plasma) and increasing during immune challenge, infection, or inflammatory stimuli, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant zebrafish IFN-γ1-1 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Zebrafish Interferon-γ (IFN-γ) is a type II interferon cytokine that plays an important role in cell-mediated immunity and regulation of antimicrobial and inflammatory responses in zebrafish (Danio rerio). IFN-γ is produced primarily by activated T lymphocytes and natural killer (NK)-like cells following stimulation by intracellular pathogens or immune signaling molecules such as IL-12 and IL-18. Zebrafish IFN-γ binds to interferon-γ receptors expressed on various immune and non-immune cells, activating intracellular signaling through the JAK–STAT pathway, particularly STAT1, which induces expression of interferon-stimulated genes involved in antigen presentation, macrophage activation, and antimicrobial defense. In zebrafish, IFN-γ contributes to activation of macrophages, enhancement of antigen presentation pathways, and control of intracellular pathogens including bacteria and parasites. Zebrafish IFN-γ is widely studied in aquatic immunology and vertebrate disease models; for example, zebrafish models of Mycobacterium marinum infection use IFN-γ expression as a key marker of host immune responses during granuloma formation and mycobacterial pathogenesis.
