  • The Zebrafish IL-17A/F1 (Interleukin-17A/F1) ELISA is designed for the quantitative measurement of IL-17A/F1 in cell culture supernatants from zebrafish (Danio rerio) and can also be used to measure IL-17A/F1 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, kidney, gill, and intestine. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-17A/F1 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-17A/F1 is a pro-inflammatory cytokine produced primarily by activated Th17-like cells and innate lymphoid cells in zebrafish and plays a central role in host defense against bacterial and fungal infections, mucosal immunity, and inflammation. In the zebrafish animal model, IL-17A/F1 is widely studied in the context of infectious disease, immune system development, inflammatory disorders, and immunomodulatory research, where its expression reflects Th17-type immune responses and inflammatory signaling. Measurement of IL-17A/F1 provides valuable insight into immune activation, cytokine signaling, disease progression, and the efficacy of therapeutic or immunomodulatory interventions, with reported levels in zebrafish typically detected at low nanogram-per-milliliter concentrations under basal conditions (~0.05-0.5 ng/mL in serum or plasma) and increasing during immune challenge, infection, or inflammatory stimulation, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant zebrafish IL-17A/F1 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Zebrafish IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays an important role in host defense and inflammatory responses in zebrafish (Danio rerio). Teleost fish possess IL-17A/F–like genes that share functional similarity with mammalian IL-17A and are expressed by lymphocyte-like cells and innate immune populations in response to microbial infection and immune stimulation. IL-17A in zebrafish acts on epithelial and stromal cells to induce expression of pro-inflammatory cytokines, chemokines, and antimicrobial peptides, promoting neutrophil recruitment and enhancing mucosal barrier immunity in gill, gut, and skin tissues. In healthy zebrafish, basal IL-17A expression is generally low but increases during bacterial, fungal, and parasitic infections, as well as following experimental inflammatory challenge. In developmental and immunology research, zebrafish IL-17A is widely studied in models of infection, intestinal inflammation, and innate–adaptive immune crosstalk, providing insight into the evolution of Th17-like responses and neutrophil-driven inflammation in vertebrates.
