  • The Ferret IL-17A (Interleukin-17A) ELISA is designed for the quantitative measurement of IL-17A in cell culture supernatants from ferrets (Mustela putorius furo) and can also be used to measure IL-17A in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lung, lymph nodes, and intestine. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-17A in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-17A is a pro-inflammatory cytokine produced primarily by activated Th17 cells and innate lymphoid cells and plays a central role in host defense against bacterial and fungal infections, mucosal immunity, and inflammatory processes. In the ferret animal model, IL-17A is widely studied in the context of respiratory infections, influenza research, inflammatory disorders, and immunomodulatory therapies, where its expression reflects Th17-mediated immune responses. Measurement of IL-17A provides valuable insight into immune activation, cytokine signaling, disease progression, and the efficacy of therapeutic or vaccine interventions, with reported levels in ferrets typically detected at low picogram-per-milliliter concentrations under basal conditions (~10-80 pg/mL in serum or plasma) and increasing to hundreds of picograms per milliliter following immune stimulation or infectious challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant ferret IL-17A protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Ferret IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays a key role in host defense and inflammatory responses in ferrets (Mustela putorius furo). IL-17A is produced primarily by Th17 cells, as well as by γδ T cells and other innate lymphoid populations, in response to microbial infection and immune stimulation. It acts on epithelial, endothelial, and stromal cells to induce expression of pro-inflammatory cytokines, chemokines (such as CXCL8), and antimicrobial peptides, thereby promoting neutrophil recruitment and strengthening mucosal barrier immunity, particularly in the respiratory and gastrointestinal tracts. In healthy ferrets, basal IL-17A expression is typically low, but levels increase during bacterial or fungal infection, viral challenge, and inflammatory stimulation. In biomedical and translational research, ferret IL-17A is of particular interest in respiratory disease models—including influenza and SARS-CoV-2—where Th17-mediated responses contribute to neutrophilic inflammation and disease severity. Characterization of IL-17A in ferrets provides valuable insight into mucosal immune regulation, host–pathogen interactions, and the balance between protective immunity and immunopathology in a widely used model of human respiratory disease.
