  • The Cynomolgus Monkey CXCL10 (C-X-C motif chemokine ligand 10, also known as IP-10) ELISA is designed for the quantitative measurement of CXCL10 in cell culture supernatants from cynomolgus monkeys (Macaca fascicularis) and can also be used to measure CXCL10 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lung, liver, and lymph nodes. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CXCL10 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. CXCL10 is a chemokine primarily produced by activated monocytes, endothelial cells, and fibroblasts and plays a central role in recruiting activated T cells, NK cells, and other immune cells to sites of infection or inflammation. In the cynomolgus monkey animal model, CXCL10 is widely studied in the context of viral and bacterial infections, autoimmune disease, immunomodulatory research, and vaccine development, where its expression reflects Th1-type immune responses and overall immune activation. Measurement of CXCL10 provides valuable insight into immune signaling, inflammation, disease progression, and the efficacy of therapeutic or vaccine interventions, with reported levels in cynomolgus monkeys typically detected at low picogram-per-milliliter concentrations under basal conditions (~20-200 pg/mL in serum or plasma) and increasing to hundreds to over a thousand picograms per milliliter during immune stimulation or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant cynomolgus monkey CXCL10 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Cynomolgus Monkey (Macaca fascicularis) CXCL10 (C-X-C motif chemokine ligand 10), also known as IP-10 (Interferon gamma–induced protein 10), is a proinflammatory chemokine belonging to the CXC chemokine family, which also includes CXCL9 (MIG) and CXCL11 (I-TAC) that regulate T-cell recruitment during antiviral and inflammatory immune responses. In cynomolgus monkeys, CXCL10 is produced by macrophages, dendritic cells, epithelial cells, endothelial cells, and fibroblasts in response to interferon-γ (IFN-γ), viral infection, or inflammatory cytokines. CXCL10 signals primarily through the chemokine receptor CXCR3, which is expressed on activated T lymphocytes, natural killer (NK) cells, and other immune cells, promoting chemotaxis and activation of these immune cells at sites of infection or inflammation. Because cynomolgus monkeys share strong genetic and immunological similarity with humans, they are widely used as preclinical animal models for infectious diseases and vaccine development, including studies of SARS-CoV-2, dengue virus, and other viral infections. In these models, CXCL10 expression is associated with interferon-driven immune responses and recruitment of CXCR3⁺ T cells to infected tissues, making cynomolgus monkey CXCL10 an important marker for studying antiviral immunity, immune-mediated inflammation, and evaluation of antiviral therapies and vaccines.
