  • The Canine Fas Ligand (FasL, also known as CD95L) ELISA is designed for the quantitative measurement of FasL in cell culture supernatants from dogs (Canis lupus familiaris) and can also be used to measure FasL in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, liver, and thymus. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring FasL in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. FasL is a transmembrane protein primarily expressed by activated T lymphocytes, natural killer (NK) cells, and some dendritic cells, and it plays a central role in inducing apoptosis, regulating immune responses, and maintaining peripheral tolerance. In the canine animal model, FasL is widely studied in the context of autoimmune diseases, transplant immunology, cancer research, and immunomodulatory therapy, where its expression reflects apoptosis-mediated immune regulation. The capture and detection antibodies in this assay are 100% homologous to Canis lupus dingo (dingo), Vulpes lagopus (Arctic fox), and Vulpes vulpes (red fox), making this kit highly relevant for comparative studies across these related canid species and allowing researchers to study immune regulation, apoptosis, and FasL-mediated pathways in broader translational models. Bovine Fas Ligand (FasL, CD95L, gene FASLG) is a type II transmembrane protein belonging to the tumor necrosis factor (TNF) superfamily, which includes related ligands such as TNF-α, TRAIL (TNFSF10), and CD40L (CD154) that regulate immune signaling, inflammation, and apoptosis. In cattle (Bos taurus), Fas ligand is expressed primarily on activated T lymphocytes, natural killer (NK) cells, and some immune regulatory cells, where it interacts with its receptor Fas (CD95) on target cells. Binding of FasL to Fas triggers the extrinsic apoptotic pathway, activating caspase signaling cascades that lead to programmed cell death. In bovine immunity, the Fas–FasL system plays an important role in regulating immune homeostasis by eliminating infected, damaged, or excess immune cells, helping control immune responses and prevent excessive inflammation. This pathway is also involved in cytotoxic T-cell–mediated killing of virus-infected or abnormal cells during infection. In bovine health, Fas ligand activity contributes to immune regulation during infectious diseases and inflammatory conditions, and dysregulation of Fas–FasL signaling may influence immune-mediated tissue damage or impaired immune control of infection. Because apoptosis and immune regulation are highly conserved processes, bovine Fas ligand is studied in comparative immunology and veterinary infectious disease research to better understand immune cell apoptosis, cytotoxic immune mechanisms, and host–pathogen interactions in livestock species.
