  • The Swine IL-28B (Interleukin-28B, also known as IFN-λ3) ELISA is designed for the quantitative measurement of IL-28B in cell culture supernatants from pigs (Sus scrofa) and can also be used to measure IL-28B in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as lung, spleen, liver, and lymph nodes. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-28B in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-28B is a type III interferon produced primarily by plasmacytoid dendritic cells, epithelial cells, and macrophages and plays a central role in antiviral defense, immune modulation, and regulation of mucosal immunity. In the swine animal model, IL-28B is widely studied in the context of viral infections, vaccine development, innate immune responses, and immunomodulatory research, where its expression reflects antiviral signaling and host defense mechanisms. Swine interferon lambda 3 (IFN-λ3), also known as interleukin-28B (IL-28B), is a member of the type III interferon family and functions as a key antiviral cytokine that signals through the IFNLR1 (IL-28Rα)/IL-10R2 receptor complex, activating JAK/STAT pathways and inducing interferon-stimulated genes (ISGs) that restrict viral replication primarily at epithelial surfaces. In swine, IFN-λ3 plays a critical role in mucosal immunity within the respiratory and gastrointestinal tracts, major entry points for economically significant pathogens such as porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus (SIV), porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), and porcine deltacoronavirus (PDCoV).
