  • The Rabbit IL-21 (Interleukin-21) ELISA is designed for the quantitative measurement of IL-21 in cell culture supernatants from rabbits (Oryctolagus cuniculus) and can also be used to measure IL-21 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, lung, and intestine. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-21 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-21 is a pleiotropic cytokine produced primarily by activated T follicular helper cells, Th17 cells, and natural killer T cells and plays a central role in regulating B and T cell responses, promoting antibody production, and modulating immune responses. In the rabbit animal model, IL-21 is widely studied in the context of vaccine development, immune regulation, infectious disease, and immunomodulatory research, where its expression reflects T cell-mediated adaptive immune responses. Measurement of IL-21 provides valuable insight into immune activation, cytokine signaling, disease progression, and the efficacy of therapeutic or vaccine interventions, with reported levels in rabbits typically detected at low picogram-per-milliliter concentrations under basal conditions (~15-120 pg/mL in serum or plasma) and increasing during immune challenge, infection, or immunostimulatory treatment, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant rabbit IL-21 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Rabbit IL-21 (Interleukin-21) is a cytokine of the common γ chain (γc) family that plays a central role in regulating adaptive immune responses in rabbits (Oryctolagus cuniculus). IL-21 is produced primarily by activated CD4⁺ T cells, particularly T follicular helper (Tfh) cells and Th17 cells, following antigen stimulation. It signals through the IL-21 receptor (IL-21R) in association with the common γ chain, activating downstream JAK/STAT pathways—predominantly STAT3—to regulate gene expression in B cells, T cells, and natural killer (NK) cells. IL-21 promotes B cell proliferation, plasma cell differentiation, immunoglobulin class switching, and antibody production, while enhancing CD8⁺ T cell cytotoxic function and NK cell activity. In healthy rabbits, basal IL-21 expression is generally low but increases during infection, vaccination, and immune activation. Rabbit IL-21 is particularly relevant in studies of vaccine immunogenicity, tuberculosis models, atherosclerosis research, ocular inflammation, and tumor immunology, where robust T cell–dependent antibody and cytotoxic responses are important. In biomedical research, rabbit IL-21 serves as a biomarker of T cell–dependent B cell help and humoral immunity and supports investigation of adaptive immune regulation and immunotherapeutic strategies.
