  • The Rabbit PD-L1 (Programmed Death-Ligand 1, also known as B7-H1 or CD274) ELISA is designed for the quantitative measurement of PD-L1 in cell culture supernatants from rabbits (Oryctolagus cuniculus) and can also be used to measure PD-L1 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, lymph nodes, lung, and tumor tissues. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring PD-L1 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. PD-L1 is an immune checkpoint protein expressed primarily by activated antigen-presenting cells, tumor cells, and some epithelial cells, and it plays a central role in immune regulation by inhibiting T cell activation and promoting peripheral tolerance. In the rabbit animal model, PD-L1 is widely studied in the context of cancer immunology, immune checkpoint research, infectious disease, and immunomodulatory studies, where its expression reflects immune suppression and regulatory pathways. Measurement of PD-L1 provides valuable insight into immune checkpoint signaling, disease progression, and the efficacy of immunotherapeutic interventions, with reported levels in rabbits typically detected at low picogram-per-milliliter concentrations under basal conditions (~10-150 pg/mL in serum or plasma) and increasing during immune activation, tumor development, or inflammatory stimulation, depending on sample type and experimental context. Rabbit Programmed Death-Ligand 1 (PD-L1) (CD274, also known as B7-H1) is an immune checkpoint protein that regulates T cell activation and immune tolerance in rabbits (Oryctolagus cuniculus). PD-L1 is a member of the B7 family of immunoregulatory proteins, which includes PD-L2 (CD273), CD80 (B7-1), CD86 (B7-2), ICOS-L (B7-H2), and B7-H3, molecules that control costimulatory and coinhibitory signaling during immune responses. PD-L1 is expressed on antigen-presenting cells such as macrophages and dendritic cells, as well as epithelial cells, endothelial cells, and some tumor or infected cells, and interacts with its receptor PD-1 (Programmed Death-1, CD279) on activated T lymphocytes. Binding of PD-L1 to PD-1 transmits inhibitory signals that reduce T cell proliferation, cytokine production, and cytotoxic activity, helping maintain immune homeostasis and preventing excessive inflammatory responses. In rabbits, PD-L1-mediated signaling is relevant in immune regulation during infection, inflammation, and tumor development, where increased PD-L1 expression may contribute to T cell suppression and immune evasion.
